  1. 1. Detection and Sequencing of Amplified Single Molecules

    Author : Rongqin Ke; Mats Nilsson; Ulf Landegren; Radoje Drmanac; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; padlock probes; proximity ligation assay; rolling circle amplification; circle-to-circle amplification; single molecule detection; amplified single molecules; sequencing; in situ; single cell; Molecular Biotechnology; Molekylär bioteknik;

    Abstract : Improved analytical methods provide new opportunities for both biological research and medical applications. This thesis describes several novel molecular techniques for nucleic acid and protein analysis based on detection or sequencing of amplified single molecules (ASMs). READ MORE

  2. 2. Development of Electrical Readouts for Amplified Single Molecule Detection

    Author : Camilla Russell; Mats Nilsson; Björn Högberg; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Rolling circle amplification; metallization; electrical DNA detection; magnetic nanoparticles; dielectrophoresis; boranephosphonate modified nucleotides; DNA elongation; Biokemi; Biochemistry;

    Abstract : Molecular diagnostics is a fast growing field with new technologies being developed constantly. There is a demand for more sophisticated molecular tools able to detect a multitude of molecules on a single molecule level with high specificity, able to distinguish them from other similar molecules. 3. 3. Biomolecular Analysis by Dual-Tag Microarrays and Single Molecule Amplification

    Author : Olle Ericsson; Ulf Landegren; Mats Nilsson; Edwin Southern; Uppsala universitet; []
    Keywords : Molecular medicine; Microarray; proximity ligation; Padlock probe; Rolling circle amplification; Splicing; single molecule; Molekylärmedicin;

    Abstract : Padlock probes and proximity ligation are two powerful molecular tools for detection of nucleic acids and proteins, respectively. Both methods result in the formation of DNA reporter molecules upon recognition of specific target molecules. READ MORE

  4. 4. MicroRNAs in HER2-Amplified Breast Cancer

    Author : Inga Newie; Bröstcancer-genetik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; miR-21; PAPD5; Breast Cancer; miR-4728; microRNA; ERBB2 HER2;

    Abstract : Background: Breast cancer is the most common female malignancy and the leading cause of cancer-related deaths worldwide. Targeted therapy against the main biomarkers estrogen receptor alpha (ER) and human epidermal growth factor receptor 2 (HER2/ERBB2) have greatly improved mortality rates, but ab initio or acquired therapy resistance is common. 5. 5. Identification and characterization of small molecules targeting MYC function

    Author : Qinzi Yan; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The MYC family genes (c-, N- and L-MYC) encode potent oncoproteins/transcription factors regulating fundamental cellular processes involved in cell proliferation, metabolism and survival, and they play an important role in tumor development. Overexpression of MYC often induce apoptosis as a failsafe mechanism to prevent tumor development and it is known to sensitize cells to genotoxic agents that induce DNA damage by triggered apoptosis.
