  1. 1. BIOACTIVE ORGANOMETALLIC COMPOUNDS

    Author : Haris Bjelosevic; Centrum för analys och syntes; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; biological; kinetic; gold; platinum; bioactive; phosphine; ruthenocene; Ferrocene;

    Abstract : Various metal complexes have been used in medicine traditionally, although their use was often connected with mystery and with a lack of empirically (clinically) proven effect. In modern times, the development of biochemistry, bioinorganic and bioorganometallic chemistry allows us more in-depth understanding of biological action of metal containing substances. READ MORE

  2. 2. Design and Synthesis of Chalcone and Chromone Derivatives as Novel Anticancer Agents

    Author : Christine Dyrager; Göteborgs universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Chalcones; Chromones; Fluorescence; Fluorophore; Cellular imaging; Anticancer; Tubulin; Microtubule; Kinase inhibitors; p38; MEK1; Palladium-mediated reactions; Molecular modeling; Structure-Activity Relationships.;

    Abstract : This thesis comprises the design and synthesis of chalcone and chromone derivatives and their use in various biological applications, particularly as anticancer agents (targeting proteins associated with cancer pathogenesis) and as potential fluorophores for live-cell imaging. Conveniently, all structures presented were synthesized from commercially available 2 ́-hydroxyacetophenones. READ MORE

  3. 3. Integrating Efficacy and Toxicity in Preclinical Anticancer Drug Development : Methods and Applications

    Author : Caroline Haglund; Elin Lindhagen; Rolf Larsson; Anna Åleskog; Sigurd Vitols; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Anticancer drugs; In vitro assays; Toxicity testing; Haematological toxicity; Primary tumour cells; Species difference; Clinical pharmacology; Klinisk farmakologi; Clinical Pharmacology; Klinisk farmakologi;

    Abstract : Preclinical testing is an important part of cancer drug development. The aim of this thesis was to establish and evaluate preclinical in vitro methods useful in the development of new anticancer drugs. In paper I, the development of non-clonogenic assays (FMCA-GM) using CD34+ stem cells for assessment of haematological toxicity was described. READ MORE

  4. 4. Interactions of Anticancer Active Platinum(II) Complexes with DNA

    Author : Johan Kjellström; Bröstcancer-genetik; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Inorganic chemistry; polyelectrolyte; medium effects; salt dependence; DSC; CD; HPLC; oligonucleotide; hairpin; DNA; anticancer active platinum II compounds; kinetics; mechanism; Oorganisk kemi; Cytology; oncology; cancerology; Cytologi; onkologi; cancer;

    Abstract : The kinetics and mechanism for interactions of the anticancer active platinum(II) complexes cis-[PtCl2(NH3)2)], cis-[PtCl2(NH3)(c-C6H11NH2)], and trans-[PtCl2-(NH3)(quinoline)] with DNA have been studied using a combination of HPLC, circular dichroism (CD), UV/vis spectroscopy, and differential scanning calorimetry (DSC). Reactions of the monoaqua forms (charge = +1) of these complexes with single-stranded DNA oligonucleotides, typically between 13-17 bases long, were found to depend on i) the composition of the reaction medium, ii) the exact nature and location of the platination target, and iii) on the DNA structure. 5. 5. Mitochondria : a target for anticancer therapy

    Author : Björn Kruspig; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Mitochondria possess a central role in several cellular metabolic pathways, maintenance of calcium homeostasis, production of reactive oxygen species (ROS) and in the regulation of various cell death modalities. A majority of cancers demonstrate aberrations in mitochondrial functions, which were shown to contribute to tumourigenesis. READ MORE
