  1. 1. Neuropeptide Y - aspects of vasomotorresponses and receptor characterization

    Author : Torun Nilsson-Bergdahl; Lund Medicin; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; vasoconstriction; RT-PCR; receptor characterization; potentiation; NPY Y1 receptor; NPY; mitogenesis; hypertension; human arteries; forearm blood flow; cell culture; antisense oligodeoxynucleotides; BIBP3226; vasodilatation.; Physiology; Fysiologi; Endocrinology; secreting systems; diabetology; Endokrinologi; sekretion; diabetologi;

    Abstract : Abstract The aims of the present thesis were to examine neuropeptide Y (NPY) responses in different human and guinea pig vascular beds and in cultured human vascular smooth muscle cells (VSMC) and to characterize, with different NPY agonists and the specific NPY Y1 receptor antagonist (BIBP3226), the NPY receptors involved. NPY-induced strong and potent contractions in human cerebral and subcutaneous arteries and in guinea pig basilar arteries. READ MORE

  2. 2. Conformationally Constrained Nucleosides, Nucleotides and Oligonucleotides : Design, Synthesis and Properties

    Author : Dmytro Honcharenko; Jyoti Chattopadhyaya; Piet Herdewijn; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Bioorganic chemistry; antisense oligonucleotides; conformationally constrained nucleosides; azetidine; aza-ENA; target affinity; RNase H; exonuclease stability; pyrene-functionalized nucleotides; fluorescence; mismatch discrimination; Bioorganisk kemi; Cell and molecular biology; Cell- och molekylärbiologi;

    Abstract : This thesis is based on six original research publications describing synthesis, structure and physicochemical and biochemical analysis of chemically modified oligonucleotides (ONs) in terms of their potential diagnostic and therapeutic applications. Synthesis of two types of bicyclic conformationally constrained nucleosides, North-East locked 1',2'-azetidine and North locked 2',4'-aza-ENA, is described. READ MORE

  3. 3. Synovial sarcoma : molecular, biological and clinical implications

    Author : Maria Törnkvist; Karolinska Institutet; Karolinska Institutet; []
    Keywords : synovial sarcoma; translocations; fusion genes; SS18-SSX; IGF-1R; cyclin D1; antisense oligodeoxynucleotides.;

    Abstract : Synovial sarcoma is a rare but mostly aggressive soft tissue tumor that mainly affects children and young adults. The tumor usually occurs in the vicinity of large joints, and may be subdivided into a classical biphasic type, a monophasic type or a poorly differentiated type, depending on histomorphological appearance. 4. 4. Gluco- and mineralocorticoid receptor regulation of regional brain neurotrophism

    Author : Anita Christiane Hansson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Neurotrophins; BDNF; NT-3; bFGF; FGF receptor; immediate early genes; c-fos; antisense oligodeoxynucleotides; adrenalectomy; stress; glucocorticoids; mineralocorticoids; glucocorticoid receptor; mineralocorticoid; receptor in situ hybridization.;

    Abstract : Gluco- and mineralocorticoid hormones exert powerful actions on brain function and are important in the mediation of the stress response and the survival of hippocampal neurons. The aim of this thesis was to study the influence of glucocorticoid (GR)/mineralocorticoid receptor (MR) actions on brain neurotrophism including analyses of immediate early genes (IEGs) and of feedback responses on GR and/or MR. 5. 5. Altered expression of contractile endothelin receptors in the vascular bed

    Author : Mikael Adner; Klinisk och experimentell allergiforskning; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cardiovascular system; upregulation; transcriptional regulation; smooth muscle cells; plasticity; organ culture; Endothelin; receptor; Kardiovaskulära systemet;

    Abstract : This thesis aims at characterizing the endothelin (ET) receptors in different vascular beds of man and rat. The ET-A receptor was shown to be the only contractile ET receptor present in all vascular regions, with the exception of the rat mesenteric vein where a weak ET-B receptor-mediated contraction was seen.
