  1. 1. Experimental Designs at the Crossroads of Drug Discovery

    Author : Ing-Marie Olsson; Svante Wold; Johan Gottfries; Michael Sjöström; Bo Nordén; Umeå universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; Chemometrics; Design of experiments; Experimental design; Multivariate data-analysis; D-optimal design; 96-well technology; Drug discovery; Assay optimisation; QSAR; Organic chemistry; Organisk kemi;

    Abstract : New techniques and approaches for organic synthesis, purification and biological testing are enabling pharmaceutical industries to produce and test increasing numbers of compounds every year. Surprisingly, this has not led to more new drugs reaching the market, prompting two questions – why is there not a better correlation between their efforts and output, and can it be improved? One possible way to make the drug discovery process more efficient is to ensure, at an early stage, that the tested compounds are diverse, representative and of high quality. READ MORE

  2. 2. Approaches to soft drug analogues of dihydrofolate reductase inhibitors : Design and synthesis

    Author : Malin Graffner Nordberg; Uppsala universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical chemistry; soft drug; dihydrofolate reductase; synthesis; Farmaceutisk kemi; Pharmaceutical chemistry; Farmaceutisk kemi; Organic Pharmaceutical Chemistry; Organisk farmaceutisk kemi;

    Abstract : The main objective of the research described in this thesis has been the design and synthesis of inhibitors of the enzyme dihydrofolate reductase (DHFR) intended for local administration and devoid of systemic side-effects. The blocking of the enzymatic activity of DHFR is a key element in the treatment of many diseases, including cancer, bacterial and protozoal infections, and also opportunistic infections associated with AIDS (Pneumocystis carinii pneumonia, PCP). 3. 3. Design, Synthesis and Thermodynamic Studies of Galectin Ligands

    Author : Maria Luisa Verteramo; Centrum för analys och syntes; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; TEKNIK OCH TEKNOLOGIER; NATURAL SCIENCES; ENGINEERING AND TECHNOLOGY; galectin; conformational entropy; solvation; thermodynamic; interaction; molecular recognition; thiodigalactoside; diastereomer; structure-based design;

    Abstract : The signaling within and between cells in biology is governed by molecular recognition between natural or synthetic ligands and proteins. This thesis project aimed to investigate the thermodynamic properties of specific interaction between synthetic ligands and galectin proteins. READ MORE

  4. 4. Drug Eluting Hydrogels : Design, Synthesis and Evaluation

    Author : Lage Ahrenstedt; Deon Bezuidenhout; Michael Malkoch; University of Cape Town; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; Drug delivery; PEGylation; Rapamycin; Dexamethasone; Controlled release; Hydrogels; Slow release;

    Abstract : Hydrogels have successfully proved themselves useful for drug delivery applications and several delivery routes have been developed over the years. The particular interest in this work was to design, synthesise and evaluate in situ forming drug eluting hydrogels, which have the potential to ameliorate the healing of cardiovascular diseases. 5. 5. Prediction, Design and Determination of Protein Structures

    Author : Sebastian Rämisch; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; protein design; Rosetta; Fold-and-Dock; leucine rich repeat proteins; coiled-coils; molecular replacement; ab initio phasing; temperature stability; fluorescent proteins;

    Abstract : The three-dimensional structure of protein is encoded in its amino acid sequence. Modern structure prediction algorithms make it possible to predict the structure of small proteins using sequence information alone. READ MORE
