  1. 1. Blood-Brain Barrier Transport : Investigation of Active Efflux using Positron Emission Tomography and Modelling Studies

    Author : Stina Syvänen; Margareta Hammarlund-Udenaes; Mats Bergström; Gunnar Blomquist; Jashvant Unadkat; Uppsala universitet; []
    Keywords : Pharmaceutical biosciences; pharmacokinetics; P-glycoprotein; blood-brain barrier; modelling; PET; active efflux; species differences; [11C]verapamil; drug development; Farmaceutisk biovetenskap;

    Abstract : This thesis examines the transport of exogenous molecules across the blood-brain barrier (BBB), focusing on active efflux, using positron emission tomography (PET), computer simulation and modelling. P-glycoprotein (P-gp) inhibition was studied using [11C]verapamil and [11C]hydroxyurea was investigated as a new marker for active efflux transport. READ MORE

  2. 2. Glutathione in the brain. Studies on efflux and possible implications in neurotoxicity

    Author : Camilla Wallin; Göteborgs universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Glutathione; excitotoxicity; hypoxia-ischemia; brain; rat; anoxia-aglycemia; g-glutamyl transpeptidase; glutamate; NMDA; oxidative stress; mitochondria; amino acids;

    Abstract : Neuronal cell death in acute insults such as stroke, trauma and epilepsy involves overactivation of glutamate receptors, calcium influx and increased formation of potentially toxic reactive oxygen species. Glutathione is a key component in the antioxidant defense system against reactive oxygen species. READ MORE

  3. 3. Stereoselective Transport of Drugs Across the Blood-Brain Barrier (BBB) In Vivo and In Vitro : Pharmacokinetic and Pharmacodynamic Studies of the (S)- and (R)-Enantiomers of Different 5-HT1A Receptor Agonists and Antagonists

    Author : Hongmei Yan; Lennart Paalzow; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Neurosciences; pindolol; propranolol; 2- di-n-propylamino tetralin; racemate; enantiomer; drug distribution; brain; rat; blood-brain barrier; Neurovetenskap; Neurology; Neurologi; Psychiatry; psykiatri;

    Abstract : Delivery of drugs to the brain requires passage across the blood-brain barrier (BBB). Both for drugs already on the market and for new drugs under development, it is important to know to what extent a drug enters the CNS. Many drugs used clinically are racemic mixtures, i.e. READ MORE

  4. 4. Drug Delivery to the Brain by Liposomes : Understanding Factors Governing Delivery Outcomes In Vivo

    Author : Yang Hu; Margareta Hammarlund-Udenaes; Teresa Dalla Costa; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; nanocarrier; liposome; blood-brain barrier; brain delivery; formulation; methotrexate; diphenhydramine; microdialysis; pharmacokinetics; model-based approach; Farmaceutisk vetenskap; Pharmaceutical Science; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy;

    Abstract : The blood-brain barrier (BBB) is the primary obstacle for efficient drug delivery to the central nervous system (CNS). One promising strategy to enhance brain delivery is to utilize nanocarriers (NC), e.g., liposomes, encapsulating CNS drugs. READ MORE

  5. 5. GLIAL HEMICHANNELS: A NEW ROUTE FOR CHEMICAL COMMUNICATION IN BRAIN

    Author : Malin Stridh; Göteborgs universitet; []
    Keywords : astrocyte; antioxidant; connexin; hemichannel; glutathione; efflux; glutamate;

    Abstract : The extracellular neurochemistry determines normal brain function and the faith of neurons after insults such as stroke. This thesis concerns the effect of extracellular events related to intense neuronal stimulation and stroke, i.e. READ MORE
