  1. 1. Development of Real-Time PCR Based Methods for Detection of Viruses and Virus Antibodies

    Author : Amal Elfaitouri; Jonas Blomberg; Helene Norder; Uppsala universitet; []
    Keywords : Microbiology; Real-time PCR; broadly targeted PCR; degenerated PCR; rTth DNA polymerase; thermolabile UNG; QPIA; IgM; Mikrobiologi;

    Abstract : Quantitative real-time PCR (QPCR) technology has been very useful for diagnosis of viral diseases. QPCR has recently reached a level of sensitivity, simplicity, and reproducibility which allows a large number of samples to be screened rapidly, make it a suitable tool for the clinical virology diagnostics. 2. 2. Detection and Quantification of Variable Viral RNA by Real-Time PCR Assays

    Author : Shaman Muradrasoli; Jonas Blomberg; Göran Friman; Maria Brytting; Uppsala universitet; []
    Keywords : Virology; Diagnosis of Viral RNA; QPCR; RNA virus; broadly targeted PCR; probe design strategy; Infectious diseases; Medicin; Medicine;

    Abstract : As the area of nucleic acid based technologies develops, so will our understanding of how structural variations in DNA and RNA pathogens are associated with disease. The overall goal of this thesis is the development of broadly targeted measurement techniques for variable viral RNA by Real-Time PCR (here referred to as quantitative reverse transcriptase PCR, QRT-PCR). 3. 3. Molecular Diagnosis of Common Viral Infectious Diseases Based on Real-Time PCR

    Author : Nahla Mohamed; Jonas Blomberg; Jan Fohlman; Elgh Fredrik; Uppsala universitet; []
    Keywords : Molecular biology; Microbiology; Virology; rTth DNA polymerase; UNG; QPCR; RNA virus; broadly targeted PCR; Molekylärbiologi;

    Abstract : Molecular biology has become an integral part of the diagnosis of infectious diseases. Recently, quantitative real-time PCR (QPCR) methods (often in the form of so-called TaqMan® systems) have been developed for the diagnosis of a wide range of infectious diseases; these techniques found valuable clinical application in the diagnosis and evaluation of progress and therapeutic success of viral diseases. 4. 4. Repertoire diversity and maturation of HIV-1 vaccine-induced B cell responses

    Author : Ganesh Eknath Phad; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : In the work presented in this thesis, I characterized B cell responses in rhesus macaques inoculated with HIV-1 envelope glycoprotein (Env)-based vaccines to define genetic and functional properties of the elicited antibodies for an improved understanding of Env immunogenicity. In Paper I, we investigated antibody heavy chain V gene segment expression in the overall B cell repertoire and in response to vaccination by using 2 approaches, 454-pyrosequencing and single-cell RT-PCR of bulk and sorted memory B cells, respectively. 5. 5. New molecular markers in mantle cell lymphoma : studies of cannabinoid receptors, 5-lipoxygenase and SOX11

    Author : Xiao Wang; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Mantle cell lymphoma (MCL) is a malignant B-cell lymphoma that accounts for 5-10% of all lymphomas and tends to occur in older adults with a higher incidence in males. The genetic hallmark of this neoplasm is the t(11;14) translocation which results in overexpression of nuclear cyclin D1. READ MORE
