  1. 1. Rational and Combinatorial Engineering of Affinity Proteins Towards Therapeutical Applications

    Author : Johan Seijsing; Torbjörn Gräslund; Uhlén Mathias; Inger Sandlie; KTH; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; neonatal Fc receptor; FcRn; affibody molecule; immunoglobulin G IgG ; serum albumin SA ; albumin binding domain ABD ; half-life extension; blocking; autoimmune disease; pH-dependence; GAD67; GAD-1; gene therapy; Parkinson’s disease; zinc finger based artificial transcription factor; Biotechnology; Bioteknologi;

    Abstract : Protein engineering has had an immense impact on the development of biological drugs, including replacement therapies with engineered versions of insulin or factor VIII to treat diabetes or bleeding disorders, and monoclonal antibodies to treat cancer and various other malignancies. Now, with the next generation of treatment modalities coming up, including monoclonal reagents based on alternative scaffolds, gene and cell therapies, the importance of protein engineering to tailor-make these treatments is likely to increase further. 2. 2. Structure, Mechanism, and Regulation of Sodium/Proton Exchangers

    Author : Iven Winkelmann; Drew David; Rajini Rao; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; sodium proton exchange; SLC9; structure; secondary active transport; membrane protein; Biochemistry; biokemi;

    Abstract : Sodium/proton exchangers (NHEs) are secondary active transporters that are ubiquitously found in all kingdoms of life. They facilitate the exchange of protons for sodium ions or other inorganic ions across biological membranes, regulating pH, sodium levels, and osmotic pressure. READ MORE

  3. 3. Neurogenesis from Neural Stem Cells, Ependymal Cells and Fibroblasts

    Author : Karthikeyan Devaraju; Lund Neurologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; adult neurogenesis; stroke; neural stem cells; Lnk; ependymal cells; FoxJ1; cortical reprogramming;

    Abstract : Stroke is a major cause of death and disability around the world. Stroke leads to loss of neurons and also other cells in the brain due to lack of blood supply. Currently no therapies are available to treat stroke-related disability. It has been shown that stroke leads to increased neurogenesis, birth of new neurons, within the brain. READ MORE

  4. 4. The role of ABC transporters and inflammation in drug-resistant epilepsy

    Author : Lora Deuitch Weidner; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : This thesis explores two pathologies thought to be related to drug resistance in epilepsy that may themselves be causally related: 1) overexpression of drug transporters in capillaries and 2) inflammation. With regard to the first hypothesis, overexpression of the ATP-binding cassette (ABC) transporters, P-glycoprotein (P-gp) and breast cancer resistance protein (BCRP), at the blood-brain barrier are thought to contribute to drug resistance in epilepsy. 5. 5. Pharmacological properties of radiotracers that measure p-glycoprotein function and density

    Author : Pavitra Kannan; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Energy-dependent transporters of the ATP-binding cassette (ABC) family regulate the movement of molecules across cellular membranes. Several of these transporters are expressed in the endothelial cells of brain microvessels (blood-brain barrier) to protect brain tissue from exposure to toxins in the blood. READ MORE
