  1. 1. Islet constitutive nitric oxide synthase and nitric oxide production. Modulatory effects on insulin and glucagon secretion

    Author : Björn Åkesson; Allmänmedicin och klinisk epidemiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacognosy; Pharmacological sciences; mouse; carbachol; muscarinic agonist; cyclic nucleotides; calcium-fluxes; NOS-inhibitors; in vitro and in vivo; insulin and glucagon secretion; Nitric oxide NO ; constitutive nitric oxide synthase cNOS ; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi; Endocrinology; secreting systems; diabetology; Endokrinologi; sekretion; diabetologi;

    Abstract : The aim of this thesis was to elucidate the role of nitric oxide (NO) in the transduction signalling of insulin and glucagon release by stimulating or inhibiting islet endogenous NO-production as well as by using NO donors. Insulin release stimulated by L-arginine, the substrate for constitutive NO-synthase (cNOS)-catalyzed NO-production, was increased by the selective NOS-inhibitor NG-nitro-L-arginine methyl ester (L-NAME) and suppressed by the intracellular NO donor hydroxylamine. 2. 2. Endoplasmic reticulum function in dendrites and dendritic spines of hippocampal neurons

    Author : Ai Na Ng; Sektion IV; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Endoplasmic reticulum; primary culture; dendritic spines; live cell imaging; STIM1; carbachol; store-operated calcium entry; 3; familial Alzheimer s disease; 5-dihyroxyphenyglycine;

    Abstract : In the hippocampus, dendritic spines are compartmentalized postsynaptic micro-domains in the excitatory synapse. Subsets of bigger and mushroom-shaped hippocampal dendritic spines contain tubules and cisterns of endoplasmic reticulum (ER) that are connected to the soma and dendrites of the neuron. READ MORE

  3. 3. Modulation of AMPA glutamate receptor functions in primary neuronal cultures

    Author : Gvido Cebers; Karolinska Institutet; Karolinska Institutet; []
    Keywords : glutamate; AMPA; cerebellar granule cells; cortical cells; cyclothiazide; AP-1; transcription factors; receptor desensitization; Ca2 ; neurotoxicity; 45Ca2 uptake; apoptosis; ischemia; muscarinic; carbachol;

    Abstract : Primary neuronal cultures were used to investigate the properties of AMPA receptors and functional consequences of their activation, as well as AMPA receptor desensitization and possibilities for its modulation. We established the presence and spontaneous functional activity of AMPA receptors in cerebellar granule and cortical cell cultures. READ MORE

  4. 4. Signal transduction in the brain : modulation of receptor-mediated inositol phospholipid breakdown by potassium and fluoride ions

    Author : Gunnar Tiger; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Inositol phospholipid breakdown; muscarinic receptors; potassium; fluoride; rat brain; pig brain;

    Abstract : Neurotransmitter receptor types mediating the generation of intracellular signals are of two types; ligand-gated ion channels and G protein coupled receptors. The effector enzyme phosphoinositide-specific phospholipase C (PLC) is modulated by stimulation of G protein coupled receptors, leading to an increased breakdown of inositol phospholipids ("Ptdlns breakdown"). 5. 5. Effects of ethanol on the expression of immediate-early genes in nerve cells

    Author : Wei-Qun Ding; Institutionen för laboratoriemedicin; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neuroadaptation; the central nervous system; AP-1; junD; junB; c-jun; fosB; c-fos; neuroblastoma SH-SY5Y; ethanol; immediate-early genes; Neurology; neuropsychology; neurophysiology; Neurologi; neuropsykologi; neurofysiologi;

    Abstract : Immediate-early genes of the fos and jun families encode proteins that dimerize to form a complex of activator protein 1 and regulate the expression of a number of genes in the nervous system. Ethanol has previously been shown to induce alterations in expression of fos and jun genes in the brain. READ MORE
