  1. 1. Cyclotides evolve : Studies on their natural distribution, structural diversity, and activity

    Author : Sungkyu Park; Göransson Ulf; Lars Bohlin; Mohamed Marahiel; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; cyclotide; cyclic cystine knot; evolution; peptide synthesis; chemical ligation; QSAR; Viola; Violaceae; phylogeny; Farmakognosi; Pharmacognosy;

    Abstract : The cyclotides are a family of naturally occurring peptides characterized by cyclic cystine knot (CCK) structural motif, which comprises a cyclic head-to-tail backbone featuring six conserved cysteine residues that form three disulfide bonds. This unique structural motif makes cyclotides exceptionally resistant to chemical, thermal and enzymatic degradation. READ MORE

  2. 2. Chemical Engineering of Small Affinity Proteins

    Author : Joel Lindgren; Amelie Eriksson Karlström; Lars Baltzer; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Affibody molecules; albumin binding domain; ligation; protein synthesis; solid phase peptide synthesis; Bioteknologi; Biotechnology;

    Abstract : Small robust affinity proteins have shown great potential for use in therapy, in vivo diagnostics, and various biotechnological applications. However, the affinity proteins often need to be modified or functionalized to be successful in many of these applications. READ MORE

  3. 3. Bottom-up Fabrication of Functional DNA Nanostructures

    Author : Erik Lundberg; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; click chemistry; gel electrophoresis; self-assembly; cross-linking; lipid membrane; fixation; linear dichroism; nanotechnology; spectroscopy; fret; dna; triplex recognition;

    Abstract : This thesis demonstrates bottom-up fabrication of a fully addressable non-repetitive network on the nanometer scale, assembled by synthetic DNA molecules. Each side constitutes a unique sequence of 10 bases, i.e. 3. READ MORE

  4. 4. Photochemical Ligation Techniques for Carbohydrate Biosensors and Protein Interaction Studies

    Author : Oscar Norberg; Olof Ramström; Stéphane Vincent; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Carbohydrates; Biosensors; CuAAC; Photochemistry; Thiol-ene yne; Perfluorophenylazide PFPA ; Lectins; Molecular recognition; Quartz crystal microbalance QCM ; Surface plasmon resonance SPR ;

    Abstract : This thesis concerns the development of surface ligation techniques for the preparation of carbohydrate biosensors. Several methodologies were developed based on efficient photochemical insertion reactions which quickly functionalize polymeric materials, with either carbohydrates or functional groups such as alkynes or alkenes. 5. 5. Theoretical studies of blue copper proteins

    Author : Mats H M Olsson; Beräkningskemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; quantum chemistry; metal-thiolate bonds; axial ligation; electron transfer proteins; reduction potential; reorganisation energy; Cu II ; Cu I ; strain; Induced-rack theory; Entatic state theory; cytochrome c oxidase; CuA; nitrite reductase; azurin; Blue copper proteins; plastocyanin; B3LYP; CASPT2; Soil Science; agricultural hydrology; Lantbrukshydrologi; marklära;

    Abstract : In this thesis, a critical investigation is presented about the role of mechanical strain for the electron-transfer properties of the blue copper proteins. It has been found that the structure of a realistic model complex of the oxidised blue-copper active site, optimised in vacuum with the B3LYP method, has the same trigonal cupric geometry as in the protein.
