21. Expression and function of ribozymes in transgenic mice
Abstract : Hammerhead and hairpin ribozymes are short RNAs that act as sequence-specific endoribonucleases by cleaving a target RNA after hybridization to complementary sequences. By changing the specific target hybridization sequence of the ribozyme, virtually any RNA can be targeted, making ribozymes a versatile tool for gene down-regulation (knockdown). READ MORE

22. Preclinical studies of ribozyme-mediated gene therapy for HIV-1
Abstract : Approximately 40 million people are infected with HIV-1 and AIDS is the number one cause of death in Africa as well as, the fourth leading cause of death globally. Patients in developed countries generally receive highly active antiretroviral therapy (HAART), even though the drugs are expensive and sometimes cause side-effects and treatment failure. READ MORE

23. Mechanistic investigation of platinum and palladium catalysed cross- coupling reactions. Implications for the Stille and Heck reactions
Abstract : The present thesis deals with various types of palladium(II) and platinum(II) complexes and their use in homogeneous catalysis. The focus has been devoted to mechanistic investigations concerning the Stille and Heck reactions and studies under both catalytic and stoichiometric conditions were conducted. READ MORE

24. Radical Cyclization Approaches to Pyrrolidines
Abstract : Five-membered rings are readily prepared by 5-exo-trig radical cyclization. This thesis is concerned with novel methodology for pyrrolidine synthesis. We have synthesised selenium containing radical precursors from aziridines and α-phenylseleno ketones, and cyclized them to 2,4- and 3,4-disubstituted pyrrolidines. READ MORE

25. Reactions of Model and Anticancer Active Platinum(IV/II) Complexes - A Kinetic and Mechanistic Study
Abstract : In this thesis reductions of model and anticancer active Pt(IV) complexes with thiols and ascorbic acid and substitution reactions of cisplatin and model Pt(II) complexes with S-donor nucleophiles (rescue agents) have been investigated using stopped-flow and UV/VIS spectrophotometry. Glutathione (GSH) reduction of the anticancer active Pt(IV) compounds t- and c-[PtCl4(cha)(NH3)] (cha = cyclohexylamine), t-[PtCl4(thiazole)(NH3)] and c-[PtCl4(NH3)2] at 25 oC was observed to be very fast.
