    Abstract : The influence of trauma (acute pancreatitis) or total parenteral nutrition (TPN) on pancreatic islet hormone secretion in relation to islet expression of inducible nitric oxide synthase (iNOS) was investigated. Acute pancreatitis resulted in an impaired glucose-stimulated insulin secretion (GSIS) which was found to be parallelled by a marked expression of iNOS and an exaggerated NO production in the pancreatic islets.     Abstract : The aim of this thesis was to elucidate the role of nitric oxide (NO) in the transduction signalling of insulin and glucagon release by stimulating or inhibiting islet endogenous NO-production as well as by using NO donors. Insulin release stimulated by L-arginine, the substrate for constitutive NO-synthase (cNOS)-catalyzed NO-production, was increased by the selective NOS-inhibitor NG-nitro-L-arginine methyl ester (L-NAME) and suppressed by the intracellular NO donor hydroxylamine.     Abstract : During the last decade it has been shown that nitric oxide (NO) and carbon monoxide (CO) are produced in our own body, and that they constitute a novel class of messenger molecules. In 1992 a constitutive NO-producing NO-synthase (cNOS) was found in the islets of Langerhans, and in 1997 we could show for the first time that the islets also contained a CO-producing constitutive heme oxygenase (HO-2).
