  1. 1. Impact of autocrine factors on physiology and productivity in Trichoplusia ni serum-free cultures

    Author : Ulrika Eriksson; Lena Häggström; Per Berglund; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biotechnology; Trichoplusia ni; High Five; insect cells; BEVS; serum-free medium; yeast extract; conditioned medium; autocrine regulation of proliferation; growth factors; cell cycle; metalloproteinase; specific productivity; Bioteknik; Bioengineering; Bioteknik;

    Abstract : The aim of this study was to increase the understanding of the mechanisms regulating cell proliferation and recombinant protein production in serum-free cultures of Trichoplusia ni (T. ni) insect cells.Conditioned medium (CM) was shown to contain both stimulatory and inhibitory factors (CM factors) influencing cell growth. 2. 2. Regulation of productivity in Trichoplusia ni and Spodoptera frugiperda Sf9 serum-free cultures

    Author : Karin Calles; Lena Häggström; Gunnel Dalhammar; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biotechnology; Insect cells; BEVS; Spodoptera frugiperda; Sf9; Trichoplusia ni; BTI-Tn-5B1-4; specific productivity; conditioned medium; conditioned medium factors; cell cycle; synchronization; G2 M; Bioteknik; Bioengineering; Bioteknik;

    Abstract : The aim of this work has been to characterize the effects of conditioned medium (CM) on insect cell productivity and physiology in order to get a better understanding about the mechanisms that regulate productivity in serum-free media. Two cell lines have been investigated, Spodoptera frugiperda (Sf9) and Trichoplusia ni (T. ni, BTI-Tn-5B1-4). READ MORE

  3. 3. Development of a protein-free fed-batch process for NS0 cells: studies on regulation of proliferation

    Author : Erika Spens; Lena Häggström; Jean-Louis Goergen; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NS0 myeloma cells; protein-free medium; fed-batch; conditioned medium; autocrine growth factors; abortive proliferation; protease activity; aprotinin; gp130; Bioengineering; Bioteknik;

    Abstract : The overall objective of this study was to investigate how NS0 cell proliferation is regulated in protein-free media. The hypothesis was that during the adaptation to growth factor-free media, animal cell lines start to produce their own autocrine growth factors to support proliferation, and after some time in a culture the effects of these factors are lost which results in cessation of proliferation. 4. 4. Murine B lymphocyte growth regulation : a study of host defence factors and their mimicry by Mycoplasma arginini

    Author : Erik Ruuth; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; B lymphocyte; growth; lymphokine; lymphokine-like; host-parasite; M. arginini; membrane acyl protein;

    Abstract : Immune responses are the result of extended clonal growth and maturation of specific precursors preexisting in low frequencies. Clonal expansion is partly controlled by soluble factors, thus, their identification is essential for the understanding of the immune response regulation. READ MORE

  5. 5. Effects of PGC1a-induced exercise adaptations in muscle on plasticity and recovery mechanisms in the CNS

    Author : Lars O Karlsson; Göteborgs universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; muscle; brain; exercise; transgenic; PGC-1α; FNDC5; irisin;

    Abstract : In this thesis, we sought to determine if muscle-derived exercise-induced signaling via PGC-1α muscle activation influences neuroplasticity under physiological or pathophysiological conditions. For this purpose, transgenic mice with muscle-specific overexpression of PGC-1α that display an endurance exercise muscle phenotype were evaluated in models of cranial irradiation and photothrombotic stroke, as well as in aging and in a voluntary running paradigm.
