  1. 6. Assessment of anti-diabetic effect of Vietnamese herbal drugs

    Author : Nguyen Khanh Hoa; Karolinska Institutet; Karolinska Institutet; []
    Keywords : G-protein; type 2 diabetes; phanoside; antidiabetic plant; hypoglycemic agent; insulin secretion; pancreatic islets; Gynostemma pentaphyllum; Anemarrhena asphodeloides; Angiopteris evecta;

    Abstract : The prevalence of type 2 diabetes mellitus is increasing in Vietnam as well as in other developing countries (China, Indian subcontinent, and Africa). Searching for hypoglycemic agents with origin from domestic herbals was considered as a useful way to find novel therapy of the disease. After treatment i.p. READ MORE

  2. 7. Pitfalls in β-cell ion imaging with fluorescent indicators and their use for real-time detection of somatostatin secretion

    Author : Mingyu Yang; Anders Tengholm; Enming Zhang; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; β-cells; cytoplasmic Ca²⁺; cytoplasmic pH; fluorescent indicators; pancreatic islets; R-GECO1; somatostatin; type 2 diabetes; Molekylär cellbiologi; Molecular Cellbiology;

    Abstract : Fluorescent ion indicators have become indispensable tools in cell physiology. Dye indicators can easily be loaded into most types of cells, while genetically encoded indicators are advantageous in allowing specific cellular or subcellular targeting. READ MORE

  3. 8. Palmitate-induced Apoptosis in Insulin-producing β-cells

    Author : Kristofer Thörn; Peter Bergsten; Claes Wollheim; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ER stress; apoptosis; palmitate; sphingosine-1-phosphate; ceramide; beta-cell; fatty acid; Medical cell biology; Medicinsk cellbiologi; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract : Type 2 diabetes is a disease characterized by the inability of pancreatic β-cells to secrete sufficient amounts of insulin to maintain normoglycemia. Increased levels of saturated fatty acids such as palmitate are believed to contribute to β-cell failure and the development of the disease. READ MORE

  4. 9. Islet constitutive nitric oxide synthase and nitric oxide production. Modulatory effects on insulin and glucagon secretion

    Author : Björn Åkesson; Allmänmedicin och klinisk epidemiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacognosy; Pharmacological sciences; mouse; carbachol; muscarinic agonist; cyclic nucleotides; calcium-fluxes; NOS-inhibitors; in vitro and in vivo; insulin and glucagon secretion; Nitric oxide NO ; constitutive nitric oxide synthase cNOS ; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi; Endocrinology; secreting systems; diabetology; Endokrinologi; sekretion; diabetologi;

    Abstract : The aim of this thesis was to elucidate the role of nitric oxide (NO) in the transduction signalling of insulin and glucagon release by stimulating or inhibiting islet endogenous NO-production as well as by using NO donors. Insulin release stimulated by L-arginine, the substrate for constitutive NO-synthase (cNOS)-catalyzed NO-production, was increased by the selective NOS-inhibitor NG-nitro-L-arginine methyl ester (L-NAME) and suppressed by the intracellular NO donor hydroxylamine. 5. 10. Regulation of glucagon secretion from pancreatic alpha cells

    Author : Yang De Marinis; diabetes och endokrinologi Genomik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Epac2; glucagon secretion; pancreatic alpha cells; K- ATP channel; N-type Ca2 channel; L-type Ca2 channel; GLP-1; cAMP; PKA; Adrenaline; PKC;

    Abstract : Glucagon secreted by pancreatic α-cells plays an important role in the regulation of blood glucose. In this thesis, different techniques such as electrophysiology, immunohistochemistry and hormone secretion assay were combined to explore the mechanisms by which glucagon secretion is regulated.
