  1. 1. Engineering of the Ultra-stable Cystine Knot Framework of Microproteins : Design, Chemical Synthesis and Structural Studies

    Author : Teshome Leta Aboye; Ulf Göransson; Julio A. Camarero; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cyclotides; cystine knot; protein engineering; NMR; solid phase; disulfide rich; ultra-stable; microprotein; PHARMACY; FARMACI; Medicinal Chemistry; Läkemedelskemi;

    Abstract : Ultra-stable cystine knotted microproteins, in which two disulfides and their connecting backbones form a circle that is penetrated by the third disulfide bonds, have attracted high interest due to their resistance to degradation in vitro and potential for the development of peptide drugs. This thesis gives new insights into engineering of that framework of microproteins, including approaches to their chemical synthesis, backbone engineering, structural and biological evaluations. 2. 2. Roles of intermolecular interactions in amyloid fibril formation mechanisms

    Author : Tony Werner; Chalmers tekniska högskola; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; disulfide-bonds; Atox1; α-synuclein; Aggregation; protein interactions; Parkinson’s disease; β-parvalbumin; macromolecular crowding; osmolytes; Amyloid fibrils;

    Abstract : Amyloid fibrils, a major pathological feature of several neurodegenerative disorders, are highly stable, insoluble aggregates of misfolded proteins. The formation of such aggregates involves a complex equilibrium between protein monomers, different on- and off-pathway transient oligomeric species, and amyloid fibrils. READ MORE

  3. 3. Fibromodulin and chondroadherin in connective tissue homeostasis

    Author : Viveka Tillgren; Reumatologi och molekylär skelettbiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Collagen; integrin; cell surface proteoglycan; extracellular matrix; and small leucine-rich repeat proteoglycan.;

    Abstract : Connective tissue contains a prominent extracellular matrix (ECM), which provides for physiological and mechanical properties. Intra- and inter-molecular interactions between proteins in this ECM provide for the formation of molecular networks. These interactions determine the shape of the tissue as well as cell behavior. 4. 4. Folding and retention of rotavirus glycoproteins in the endoplasmic reticulum

    Author : Ali Mirazimi; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ER; rotavirus; VP7; NSP4; oligosaccharide processing; N-linked glycosylation; protein disulfide isomerase; calnexin; ATP; ER retention;

    Abstract : The endoplasmic reticulum (ER) is a membrane bound organelle composed of a network of cisternae stretching throughout the cytoplasm. Functionally, the ER performs many essential processes, such as glycosylation, and biosynthesis of lipids; it provides a redox potential condition for forming S-S linkages, and a quality control system, and functions as an intracellular calcium store. 5. 5. Structure and Interactions of the Extracellular Matrix Protein PRELP

    Author : Eva Bengtsson; Institutionen för experimentell medicinsk vetenskap; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; reumatologi; muskelsystem; Skelett; extracellular matrix; PRELP; heparin; heparan sulfate; perlecan; collagen; cell-binding; Skeleton; rheumatology locomotion; muscle system; Biochemistry; Metabolism; Biokemi;

    Abstract : This thesis describes the structure and interactions of a novel extracellular matrix protein - PRELP (proline arginine-rich end leucine-rich repeat protein). The amino acid sequence of PRELP showed that it belonged to the leucine-rich repeat (LRR) protein family in connective tissues. READ MORE
