  1. 1. Drug Dissolution under Physiologically Relevant Conditions In Vitro and In Vivo

    Author : Eva Persson; Hans Lennernäs; Bertil Abrahamsson; Peter Langguth; Uppsala universitet; []
    Keywords : Biopharmacy; solubility; drug dissolution; food effects; poorly soluble drugs; P-gp inhibition; drug absorption; pemeability; bile acids; phospholipids; human intestinal fluid; simulated intestinal fluid; neutral lipids; solid phase extraction; HPLC; Efflux; Biofarmaci;

    Abstract : The general aim of the present project was to increase the understanding of the in vivo dissolution of poorly soluble drugs and thereby improve possibility to predict in vivo solubility from substance properties. Increased understanding of the in vivo limitations of drug solubility could potentially also generate ideas for improved formulation principles for poorly soluble compounds and more relevant in vitro dissolution test methods used in formulation development. 2. 2. Novel and refined small-scale approaches to determine the intrinsic dissolution rate of drugs

    Author : Sara B. E. Andersson; Göran Alderborn; Göran Frenning; Peter Langguth; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; dissolution; intrinsic dissolution rate; poorly water-soluble compounds; single particle dissolution; solubility; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : Many drugs are administered as crystalline particles compressed into tablets and taken orally. When the tablet reaches the gastrointestinal tract, it disintegrates and the drug particles dissolve in the gastrointestinal fluid. The dissolved molecules are absorbed across the intestinal membranes into the bloodstream to reach their target sites. READ MORE

  3. 3. Mechanistic Modelling of Drug Dissolution and Matrix Formulations: Rotating Disc Dissolution, Dissolution in a Stagnant Medium, Swelling and Dissolving Matrix Systems

    Author : Erik Kaunisto; Avdelningen för kemiteknik; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Rotating Disc; Diffusion; Dissolution; Controlled Release; Hydrophilic matrices; ESPI;

    Abstract : Popular Abstract in Swedish Under årens lopp har människan gjort stora landvinningar inom läkemedelsforskning, vilket har lett fram till avancerade läkemedel som idag tas för givna. Avancerade läkemedel sätter dock höga krav på tillverkarna av läkemedel, vilket har lett till att framtagandet av nya läkemedel är en ganska komplicerad process. READ MORE

  4. 4. Advanced Methods for Evaluation of the Performance of Complex Drug Delivery System

    Author : Caroline Alvebratt; Christel Anna Sofia Bergström; Maria Strömme; Caitriona O'Driscoll; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; amorphous solid dispersion; coadministration; digestion; dissolution; drug absorption; enabling formulation; in vitro assay; lipid-based formulation; mesoporous carrier; supersaturation; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : Low oral bioavailability of drugs originating from poor aqueous solubility is a common issue in drug development. Various enabling formulations have been presented to circumvent this limitation, many making use of supersaturation. In these, the drug is delivered to the gastro-intestinal lumen in a high energy state e.g. READ MORE

  5. 5. Drug absorption in the lungs : studies in the isolated perfused rat lung model combined with physiologically based biopharmaceutics modelling

    Author : Johanna Eriksson; Hans Lennernäs; Erik Sjögren; Helena Thörn; Ben Forbes; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pulmonary drug absorption; physiologically based biopharmaceutics modeling; pulmonary drug delivery; Biofarmaci; Biopharmaceutics;

    Abstract : Pulmonary delivery of drugs is the preferred route of administration for treatment of local lung diseases like asthma and chronic obstructive pulmonary disease. Recently, there has also been increased interest in systemic delivery of drugs via the lungs to avoid problems with low and/or variable gastrointestinal absorption, and as a needle-free alternative for drugs that cannot be ingested.
