  1. 16. Drug Delivery to the Brain by Liposomes : Understanding Factors Governing Delivery Outcomes In Vivo

    Author : Yang Hu; Margareta Hammarlund-Udenaes; Teresa Dalla Costa; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; nanocarrier; liposome; blood-brain barrier; brain delivery; formulation; methotrexate; diphenhydramine; microdialysis; pharmacokinetics; model-based approach; Farmaceutisk vetenskap; Pharmaceutical Science; Farmakokinetik och läkemedelsterapi; Pharmacokinetics and Drug Therapy;

    Abstract : The blood-brain barrier (BBB) is the primary obstacle for efficient drug delivery to the central nervous system (CNS). One promising strategy to enhance brain delivery is to utilize nanocarriers (NC), e.g., liposomes, encapsulating CNS drugs. READ MORE

  2. 17. Development and application of new methods for anticancer drug discovery and early evaluation

    Author : Sumeer Dhar; Uppsala universitet; []
    Keywords : Medical sciences; Drug development; tumor cell lines; cytotoxic drugs; neural network; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; klinisk farmakologi; Clinical Pharmacology;

    Abstract : A non-clonogenic assay using the redox dye Alamar Blue and primary human tumor cells (PHTC) from patients and human tumor cell lines (HTCL) was established for rapid screening and evaluation of anticancer drugs. PHTC cultures from chronic lymphocytic leukemia (CLL) and ovarian carcinoma (Ovca) were compared with renal carcinoma (ACHN) and lymphocytic (CCRF-CEM) HTLCs in response to 63 toxic or non-toxic compounds. 3. 18. Developmental Aspects of Drug Transport Across the Blood-Brain Barrier

    Author : Jörgen Bengtsson; Margareta Hammarlund-Udenaes; Ann-Cathrin Engwall; Sven Björkman; Elizabeth C.M. de Lange; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Blood-brain barrier; development; active transport; tight junction proteins; microdialysis; recovery; morphine; nitrofurantoin; Bcrp; microarray; real-time PCR; in vitro; in vivo; LC-MS MS; Biopharmacy; Biofarmaci; Pharmacokinetics and Drug Therapy; Farmakokinetik och läkemedelsterapi;

    Abstract : The developmental aspect of drug transport across the blood-brain barrier (BBB) was investigated. Microdialysis was used to study unbound morphine BBB transport at different ages in sheep. An in vitro study was performed to find differentially expressed genes in brain capillary-rich fractions of the brain in rats of different ages. READ MORE

  4. 19. Intestinal barriers to oral drug absorption: Cytochrome P450 3A and ABC-transport proteins

    Author : Helena Engman; Per Artursson; Hans Lenernäs; Heyo K Kroemer; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutics; oral drug delivery; bioavailability; human jejunum; Caco-2; ABC-transporter; P-glycoprotein; CYP3A; Galenisk farmaci; Pharmaceutics; Galenisk farmaci;

    Abstract : The subject of this thesis was to study two intestinal barriers to oral drug bioavailability, drug efflux proteins of the ABC-transporter family, and in particular ABCB1/P-glycoprotein (Pgp), and the drug metabolizing enzyme cytochrome P450 (CYP) 3A4. At the onset of this thesis, similarities between CYP3A4 and Pgp in terms of their tissue distribution and gene regulation, along with overlapping substrate specificities, had generated the hypothesis that CYP3A4 and Pgp may have a complementary function and thus form a coordinated intestinal barrier to drug absorption and gut wall metabolism. 5. 20. Advanced Mass Spectrometry Imaging in Neuropharmacology

    Author : Theodosia Vallianatou; Per E. Andrén; Anna Nilsson; Mohammadreza Shariatgorji; Maria Karlgren; Irena Loryan; Nathalie Agar; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; mass spectrometry imaging; neuropharmacology; blood brain barrier; drug-drug interactions; aging; tacrine;

    Abstract : Mass spectrometry imaging (MSI) has emerged as a valuable approach for mapping multiple molecular species in sections of diverse tissues. It enables simultaneous detection of numerous compounds (from neurotransmitters to small proteins) in the brain at relatively high lateral resolution (>5 μm) on a routine basis. READ MORE
