  1. 1. Homing Endonucleases and Horizontal Gene Transfer in Bacteria and Bacteriophages

    Author : David Nord; Britt-Marie Sjöberg; Steinar Johansen; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; dsDNA cleavage; ribonucleotide reductase; GIY-YIG motif; Molecular biology; Molekylärbiologi; molekylärbiologi; Molecular Biology;

    Abstract : Homing endonuclease genes (HEGs) are selfish genetic elements that mediate their own super-Mendelian inheritance. This is mediated by the homing endonuclease cleavage of a HEG- allele followed by recombination-repair with a HEG+ allele.The majority of the HEGs are encoded in intervening sequences (IVSs). 2. 2. Cationic Interactions with Nucleic Acids With Focus on Anticancer Active Platinum(II) Complexes

    Author : Åse Sykfont Snygg; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; in-line cleavage; Cisplatin; oxaliplatin; circular dichroism; DENSPM; polyamines; siRNA; pseudoknot; RNA; thermal melting; anticancer active Pt II complex; DNA;

    Abstract : It is well documented that the antitumour activity of the Pt(II)-complex cisplatin, and similar anticancer active complexes, is a result of metalation of DNA, and thereby inhibition of cell proliferation. Since nucleic acids are polyanions at physiological pH, the rate of adduct formation with positively charged complex, as the hydrolyzed Pt(II) complexes studied in this work, is influenced by the ionic environment surrounding the polymer. 3. 3. Studies on nucleic acids chemistry

    Author : Nitin Puri; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Bioorganic chemistry; Bioorganisk kemi; Bioorganic chemistry; Bioorganisk kemi; bioorganisk kemi; Biorganic Chemistry;

    Abstract : This thesis is divided into three parts: Part 1: Synthesis and conformational analysis of cyclic RNAs, and comparison with lariat RNAs modelling the lariat formed during processing of Group II and Nuclear pre-mRNA. Synthesis of a hexameric lariat RNA model with a tetranucleotidyl "2' → 5' loop" and a dinucleotidyl "3'-tail" has been accomplished. READ MORE

  4. 4. Development of benzoquinoquinoxaline derivatives as triplex-specific probes : Recognition of DNA structures at repeats sequences

    Author : Helen Bergquist; Rula Zain; Keith Fox; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Triplex; H-DNA; BQQ; BQQ-OP; BQQ-Bodipy; triplet repeat; DNA; non-B-DNA; pkd1; frataxin; Friedreich s ataxia; Molecular biology; Molekylärbiologi; Molecular Biology; molekylärbiologi;

    Abstract : Repeat sequences are associated with several human diseases, such as Friedreich’s ataxia, polycystic kidney disease and cancer. These sequences can form non-B-DNA structures, including triplex (H-DNA) DNA, and are associated with genomic instability and altered gene expression. READ MORE

  5. 5. Molecular mechanisms of mammalian mitochondrial DNA replication and transcription

    Author : Mina Pellegrini; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Mitochondrion; DNA polymerase gamma; mtSSB; TWINKLE; DNA replication; transcription; mTERF; MTERF3; MTERF2; nucleoid;

    Abstract : Mitochondria are cytoplasmic organelles in eukaryotic cells, mainly devoted to the synthesis of ATP, through the Oxydative Phospshorylation System (OXPHOS). The organelle has its own genome, the mitochondrial DNA (mtDNA) that encodes for all the RNA components of the mitochondrial translation apparatus (two rRNAs and 22 tRNAs) and for 13 subunits of the OXPHOS complexes.
