  1. 1. Fragment-based drug discovery : Novel methods and strategies for identifying and evolving fragment leads

    Author : Edward A. FitzGerald; Helena Danielson; Hanna-Kirsti Schrøder Leiros; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Drug Discovery; Biophysics; Fragment-based drug discovery; Epigenetics; Biosensors; Surface Plasmon Resonance; Interaction Analysis; Second-Harmonics; Biokemi; Biochemistry;

    Abstract : The need for new drugs became ever more apparent in the year 2020 when the world was faced with a viral pandemic. How drugs are discovered and their relevance to society became part of daily discussions in workplaces and homes throughout the world. Consequently, efficient strategies for preclinical drug discovery are clearly needed. READ MORE

  2. 2. Drug discovery against leishmaniasis : Bio- and chemoinformatic guided strategies for target evaluation and hit identification

    Author : Elisabet Vikeved; UCM Alsmark; Christian Sköld; David Horn; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Leishmaniasis; Drug discovery; Lateral gene transfer; Comparative genomics; Virtual screening; Target fishing; Marine natural products;

    Abstract : Leishmaniasis is a neglected tropical disease mainly affecting poor people in developing countries. It is caused by infections of flagellated protozoa belonging to genus Leishmania. The few available drugs are associated with problems such as low effectiveness, severe side effects and resistance development. 3. 3. Structural studies of drug targets and a drug metabolizing enzyme

    Author : Daniela Cederfelt; U. Helena Danielson; Doreen Dobritzsch; Inari Kursula; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Biophysics; Protein structure; X-ray crystallography; Cryogenic electron microscopy; Enzymology; Drug discovery; Pyrimidine degradation; Biokemi; Biochemistry;

    Abstract : The work presented in this thesis describes how structural information about a protein can be acquired, and how it can be used to answer scientific questions about proteins’ function, their dynamic behaviour and their interactions with other proteins or ligands.The catalytic function of the pyrimidine-degrading, drug metabolizing enzyme β-ureidopropionase (βUP) is dependent on the shift between oligomeric states. 4. 4. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development

    Author : Matthis Geitmann; U. Helena Danielson; Marc H. V. van Regenmortel; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; SPR biosensor; HIV-1 reverse transcriptase; HCMV protease; interaction kinetics; drug discovery; non-nucleoside inhibitor; resistance; Biokemi; Biochemistry; Biokemi;

    Abstract : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 5. 5. Dynamic Sulfur Chemistry : Screening, Evaluation and Catalysis

    Author : Rémi Caraballo; Olof Ramström; Nicolas Giuseppone; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Dynamic combinatorial chemistry; dynamic sulfur chemistry; thioglycoside; mutarotation; disulfide metathesis; hemithioacetal formation; 1H STD-NMR; inhibition; phosphine; Concanavalin A; beta-galactosidase; dynamic drug discovery.; Chemistry; Kemi; Organic chemistry; Organisk kemi; Bioorganic chemistry; Bioorganisk kemi;

    Abstract : This thesis deals with the design, formation and evaluation of dynamic systems constructed by means of sulfur-containing reversible reactions, in organic and aqueous media and under mild conditions. In a first part, the synthesis of thioglycoside derivatives, constituting the biologically relevant starting components of the dynamic systems, is described. READ MORE
