  1. 1. Automating STED microscopy for functional and structural live-cell imaging

    Author : Jonatan Alvelid; Ilaria Testa; Eggeling Christian; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; STED; microscopy; nanoscopy; super-resolution microscopy; automation; image analysis; fluorophores; Biologisk fysik; Biological Physics;

    Abstract : Optical microscopy imaging methods are today invaluable tools for studies in life sciences as they allow visualization of biological systems, tissues, cells, and sub-cellular compartments from millimetres down to nanometres. The invention and development of nanoscopy in the past 20 years has pushed fluorescence microscopy down to the nanoscale, reaching beyond the natural diffraction limit of light that does not allow focusing of visible light below sizes of around 200 nm, and into the realm of what was previously only thought possible with electron microscopy. 2. 2. Characterization of Fluorescent Dyes for Biochemical Applications. Chemometric Analysis of Spectroscopic Data

    Author : Jan Nygren; Chalmers tekniska högskola; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; principal component analysis; fluorescein; chemometrics; optical spectroscopy; fluorophores; thiazole orange;

    Abstract : Fluorescent dyes are used in a number of different biochemical applications to visualize and probe biomolecules. The aim of this work was to characterize the spectroscopic properties of two such dyes, fluorescein and thiazole orange, and to develop chemometric methods to extract further information from these data. 3. 3. Fluorescent Probes and Protein Misfolding: Methods and Applications

    Author : Irem Nasir; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Protein misfolding; fluorophores; filter-trap; site-specific fluorophore labeling; amyloid-beta; microscopic rate constants; solvatochromic probes; alpha-synuclein; lipid vesicles; nanoparticles; protein adsorption;

    Abstract : Protein misfolding is a frequently occurring event in living cells and without the quality control, the consequences are disasterous. In this thesis, we studied protein misfolding in vitro by employing a variety of fluorophores, to either detect the highly ordered aggregates of intrinsically disordered proteins/peptides or slight misfolding of a natively folded protein. 4. 4. Organic Azides : Functional Molecules and Materials

    Author : Giampiero Proietti; Peter Dinér; Per I. Arvidsson; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; perfluorinated Aromatic Azide; Photoactivatable; fluorophores; aggregation-induced emission; sulfonimidamides; sulfoximes; chiral auxilary; organogel; azides reduction; Ni-B; Kemi; Chemistry;

    Abstract : The work presented in this thesis stems from the chemistry of the azido group, and more specifically from the unique reactivity of perfluorinated aromatic azides and how to use this reactivity to access new types of molecules to enable new applications in asymmetric synthesis and materials.In the first section of this thesis, a photoactivatable fluorescence probe is presented, where the non-luminescent azide was activated via a UV-light-promoted intramolecular N–H insertion reaction forming a fluorescence emitter. 5. 5. 3D super-resolution microscopy of living cells using reversibly switchable fluorophores

    Author : Andreas Bodén; Ilaria Testa; Francisco Balzarotti; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; RESOLFT; 3D imaging; super-resolution; microscopy; reversibly switchable fluorescent proteins; Biological Physics; Biologisk fysik;

    Abstract : Traditional optical microscopy techniques are limited in spatial resolution due to the wave nature of light. This means that neighboring objects separated by a distance smaller than about 200 nm cannot be distinguished. READ MORE
