  1. 1. Glutathione transferases : molecular cloning, site-directed mutagenesis and structure-function studies

    Author : Gun Stenberg; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : Three distinct glutathione transferases (GST) cDNA clones were obtained by screening of tumour cell line cDNA libraries. The cDNA clones were expressed in Escherichia coli and the enzymes characterized. Rat GST 8-8 is the only member in the multifunctional GST family for which a specific cellular function has been proposed. READ MORE

  2. 2. Glutathione reductase from erythrocytes : purification, characterization and identification of functional histidine and arginine residues of the enzyme

    Author : Vijayakumar Boggaram; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : This investigation aims at the development of a rapid, simple purification procedure for glutathione reductase and at an exploration of the chemical groups of the enzyme essential for activity.Glutathione reductase from porcine erythrocytes was purified to homogeneity by the use of affinity chromatography on 2’,5’ ADP-Sepharose 4B. 3. 3. Evolutionary Analysis and Posttranslational Chemical Modifications in Protein Redesign : A Study on Mu Class Glutathione Transferases

    Author : Ylva Ivarsson; Bengt Mannervik; Gun Stenberg; Richard N. Armstrong; Uppsala universitet; []
    Keywords : Biochemistry; glutathione transferase; evolutionary analysis; positive selection; epoxide; stereoselectivity; protein modification; Biokemi;

    Abstract : Glutathione transferases (GSTs) constitute a family of multifarious enzymes that conjugate glutathione (GSH) with a wide range of electrophiles. GSTs are grouped into different classes based on protein sequence similarities. Despite high sequence identities between GSTs of the same class they often display different substrate specificites. 4. 4. Studies on hepatic glutathione transferases from rat and man : 1. A kinetic mechanism for glutathione transferase A from rat liver. 2. Transferase µ, a new form of glutathione transferase in human liver

    Author : Margareta Warholm; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : This study of glutathione transferases was undertaken to achieve a better understanding of the catalytic and molecular properties of various isozymes of this group of drug-metabolizing enzymes.The steady-state kinetics of purified glutathione transferase A from rat liver with glutathione (GSH) and 1,2-dichloro- 4-nitrobenzene as substrates, did not follow a simple Michaelis- Menten equation. 5. 5. The Quest for Functional Quasi-Species in Glutathione Transferase Libraries

    Author : Arna Rúnarsdóttir; Bengt Mannervik; Ralf Morgenstern; Uppsala universitet; []
    Keywords : glutathione transferase; directed evolution; multivariate analysis; quasi-species; isothiocyanates; Biokemi; Biochemistry;

    Abstract : Glutathione transferases (GSTs) are good candidates for investigations of enzyme evolution, due to their broad substrate specificities and structural homology. The primary role of GSTs is to act as phase II detoxifying enzymes protecting the cell from toxic compounds of both endo- and exogenous origins. READ MORE
