  1. 1. Heterologous expression and characterization of xyloglucan endotransglycosylases from Populus tremula x tremuloides

    Author : Åsa Kallas; KTH; []
    Keywords : xyloglucan endotransglycosylase; transglycosylation; hybrid aspen; cell expansion; heterologous expression; Pichia pastoris; N-glycosylation; X-ray crystallography;

    Abstract : Xyloglucan endotransglycosylases (XETs) are enzymes involvedin the reorganization of the primary cell wall. This thesisdescribes several aspects of XET16A from hybrid aspen (Populustremula x tremuloides). The gene was isolated from the hybridaspen cDNA library and the protein was expressed heterologouslyin the methylotrophic yeast Pichia pastoris. READ MORE

  2. 2. Bench-Scale Production of Heterologous Proteins from Extremophiles- Escherichia coli and Pichia pastoris based expression systems

    Author : Santosh Ramchuran; Bioteknik; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Glycoside hydrolase; Heterologous protein; High cell density cultivation Expression systems; Bioteknik; Biotechnology; Fed-batch;

    Abstract : Over the past few years considerable research attention has been assigned to extremophiles as sources of extremozymes due to their applicability in industrial processes, and the development of eco-friendly technologies. The establishment of efficient production strategies for heterologous proteins is an empirical process requiring broad background knowledge on available expression systems together with their major advantages and shortcomings. 3. 3. Laccases and Oxalate-Degrading Enzymes Heterologous Expression and Novel Applications in Lignocellulose Processing

    Author : Pierre Cassland; Teknisk mikrobiologi; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; virology; mycology; Mikrobiologi; virologi; mykologi; bacteriology; Microbiology; Lignocellulose Processing; Heterologous expression; Laccases; Oxalate-Degrading Enzymes; bakteriologi;

    Abstract : Lignocellulose constitutes a vast renewable resource for the production of, for example, paper and bioethanol. The potential of using laccase and oxalate-degrading enzymes in novel applications for the processing of lignocellulose was investigated in this work. READ MORE

  4. 4. Engineering yeast for improved recombinant protein production

    Author : Yanyan Wang; Chalmers tekniska högskola; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; NATURVETENSKAP; NATURAL SCIENCES; point mutation; genome-scale modeling; recombinant protein; omics analysis; CRISPR Cas9; protein secretion; Saccharomyces cerevisiae;

    Abstract : Recombinant proteins are broadly used from basic research to therapeutic development and include industrial enzymes and pharmaceutical proteins. The increasing demand for improved production and enhanced quality of recombinant proteins requires robust biotech-based strategies to overcome the limitations of protein extraction from natural sources. 5. 5. Protein production, characterization and structure determination in structural genomics

    Author : Esmeralda A. Woestenenk; Torleif Härd; Göran Karlsson; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Biology; protein production; protein solubility; fusion tags; nuclear magnetic resonance; structure determination; Biologi; Biology; Biologi;

    Abstract : This thesis covers the process from expression of a heterologous gene in Escherichia coli to structure determination of a protein by nuclear magnetic resonance (NMR) spectroscopy. The first part concerns structural genomics-related parallel screening studies on the effect of fusion tags (in particular the His tag) on protein solubility and the use of fusion tags in fast, parallel purification protocols intended for initial biophysical characterization of human proteins produced in E.
