  1. 1. Anthracyclines : Toxicity and chemotherapy-induced mucositis

    Author : Fredrik Kullenberg; Hans Lennernäs; Erik Sjögren; David Dahlgren; Femke Heindryckx; Henrik Rönnberg; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Anthracycline; doxorubicin; idarubicin; toxicity; intracellular concentration; IC50; chemotherapy-induced mucositis; apoptosis; proliferation; permeability; anti-inflammatory drugs; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : Anthracyclines belong to a class of cytostatic compounds that are commonly used to treat various types of cancers, including lymphoma, breast cancer and primary liver cancer. The aim of this thesis was to study how two anthracyclines, doxorubicin and idarubicin, induced cytotoxicity in tumor cells, as well as their off-target effect on the gastrointestinal system. 3. 3. On mechanisms of drug resistance in acute myeloid leukemia

    Author : Malin Prenkert; Ulf Tidefelt; Hilja Strid; Mats Merup; Örebro universitet; []
    Keywords : MEDICINE; MEDICIN; Biomedicin; Biomedicine;

    Abstract : In this thesis focus has been to increase the knowledge and understanding of some of the mechanisms responsible for drug resistance in acute myeloid leukemia, as well as identify possibilities to predict drug resistance at diagnosis.We have studied the intracellular behavior of cytostatic drugs and their main metabolites (paper I) and the cellular response to cytostatic drugs (paper III). 4. 4. In vitro studies of cytostatic drugs and growth factors on leukemic and normal hematopoietic cells : based on in vivo intracellular pharmacokinetics

    Author : Britt Sundman-Engberg; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ANLL; Normal bone marrow; in vitro; predictive tests; pharmacokinetics; cytostatic drugs; growthfactors;

    Abstract : The aims were to develop, evaluate and apply in vitro methods for cytostatic drug sensitivity, based on intracellular concentrations found after in vivo infusions. The intracellular pharmacokinetics of anthraquinones and ara-C were compared during and after infusion to leukemic patients. Drug concentrations were evaluated with HPLC. READ MORE

  5. 5. Expression of multidrug resistance genes and proteins and effect of selenite in anthracycline-resistant human tumor cell lines

    Author : Kerstin Jönsson Videsäter; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Multidrug resistance; cross-resistance profile; ATP-bioluminescence assay; pglycoprotein; MRP1; selenium; thioredoxin reductase; glutathione reductase.;

    Abstract : Treatment failure due to development of drug resistance is a serious complication in cancer treatment and the major cause of death in these patients. This thesis has focused on to gain knowledge about the mechanisms of multidrug resistance and to establish in vitro methods to characterize the resistant tumor cell phenotype. READ MORE
