  1. 1. Early assessment of efficacy and safety of novel anticoagulants as exemplified by direct thrombin inhibitors. Non-clinical pharmacodynamic studies in vitro and in vivo

    Author : Margareta Elg; Göteborgs universitet; Göteborgs universitet; Gothenburg University; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; animals; antidote; bleeding; inhibition constant; inogatran; association rate constant; melagatran; thrombin inhibitor; thrombosis; fibrinolysis;

    Abstract : The drawbacks of today's anticoagulants are parenteral use (heparins), a narrow interval between efficacy and bleeding, and the high variability of effect with a consequent need for frequent monitoring (coumarins). The aim of the current studies was to evaluate properties important for efficacy and safety of novel anticoagulants, as exemplified by direct thrombin inhibitors, with non-clinical methods. 2. 2. Studies of myocardial ischaemia followed by reperfusion : New experimental models and their application

    Author : Yozo Uriuda; Karolinska Institutet; Karolinska Institutet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; myocardial infarction; myocardial ischaemia; reperfusion injury; animal experiment; rat; pig; dog; coronary venous retroinfusion; synchronised coronary venous retroperfusion; thrombosis; thrombolysis; inogatran; endothelin;

    Abstract : Studies of Myocardial Ischaemia Followed by Reperfusion New Experimental Models and their Application Yozo Uriuda Despite great efforts to reduce morbidity and mortality in ischaemic heart disease, this is still the most common cause of death in the industrialised countries. To improve the management of such patients, further knowledge is needed of the pathophysiology of ischaemic heart disease. READ MORE

  3. 3. The effect of thrombin inhibitors on coagulation activity and generation of activated protein C

    Author : Rikard Linder; Karolinska Institutet; Karolinska Institutet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP;

    Abstract : The direct thrombin inhibitor inogatran was, in the TRIM trial (n=1209), compared with unfractionated heparin (UFH) concerning the effect on cardiovascular end-points in unstable coronary artery disease. The results in TRIM demonstrated a trend towards a benefit for UFH during the treatment phase, which was reduced after drug withdrawal. READ MORE

  4. 4. Experimental studies on thrombosis and thrombolysis : With special reference to importance of lys-plasminogen, active site thrombin inhibitors and stable fish oil

    Author : Liying Chen; Uppsala universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Surgery; Thrombolysis; thrombosis; recombinant tissue-type plasminogen activator; lys- and glu-plasminogen; aspirin; thrombin inhibitors; platelets; superoxide dismutase; stable fish oil; Kirurgi; Surgery; Kirurgi; rättsmedicin; Forensic Medicine;

    Abstract : Dissolution and prevention of thrombus in the atherosclerotic coronary artery have become an important part of treatment therapy of acute myocardial infarction. However, the currently most effective thrombolytic agent, recombinant tissue-type plasminogen activator (rt-PA), only elicits 80% recanalization of thrombotically occluded coronary arteries and early occlusion occurs in about 40% of successfullyrecanalized vessels. 5. 5. Inflammation and Coagulation Activity in Unstable Coronary Artery Disease and the Influences of Thrombin Inhibition

    Author : Jonas Oldgren; Steen Husted; Uppsala universitet; []
    Keywords : Medical sciences; Unstable coronary artery disease; inflammation; coagulation; thrombin inhibition; MEDICIN OCH VÅRD; MEDICINE; MEDICIN; kardiologi; Cardiology;

    Abstract : In patients with unstable coronary artery disease, this study evaluated the degree of inflammation and coagulation activity, relations to myocardial cell damage, prognosis, and influences of randomisation to 72 h infusion with three different doses of inogatran, a direct thrombin inhibitor (n=904), or unfractionated heparin (n=305). Anticoagulant treatment effects were evaluated with aPT time. READ MORE
