  1. 1. First-pass Intestinal Metabolism of Drugs : Experiences from in vitro, in vivo and simulation studies

    Author : Helena Anna Thörn; Hans Lennernäs; Paul Alfred Dickinson; Brian Houston; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacokinetics; metabolism; CYP3A4; CYP2C9; CYP2D6; UGT; glucuronidation; physiologically based pharmacokinetic model; modelling; Biopharmaceutics; Biofarmaci;

    Abstract : The bioavailability of a drug can be described as the fraction of an orally administered dose that reaches the systemic circulation and is often limited by first-pass metabolism in the gut and the liver. It is important to have knowledge about these processes since the systemic blood drug concentration is tightly connected to the effect of the drug. READ MORE

  2. 2. Exploring Intestinal Ischemia : An experimental study

    Author : Rolf Fröjse; Conny Arnerlöv; Ola Winsö; Stefan Lehtipalo; Eric Wahlberg; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Surgery; gastrointestinal tonometry; continuous; pig; pHi; intestinal ischemia; intestinal vascular bed; lactate; tissue oxygen tension; microdialysis; dopexamine; Kirurgi; Surgery; Kirurgi; Surgery; kirurgi;

    Abstract : Background and aims: Unrecognized intestinal mucosal ischemia in severely ill patients may trigger development of multiple organ failure. Such ischemia can be evaluated by intraluminal tonometry reflecting mucosal PCO2 and intramucosal pH (pHi). READ MORE

  3. 3. Regulation of the cytoskeleton and the adhesiveness of intestinal epithelial cells by leukotriene D4

    Author : Ramin Massoumi; Institutionen för translationell medicin; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; oncology; Cytology; patologisk anatomi; Patologi allmän ; pathological anatomy; General pathology; PKC; vinculin; actin filament; focal adhesion; LTD4; integrin; cancerology; Cytologi; onkologi; cancer; Biochemistry; Metabolism; Biokemi; metabolism;

    Abstract : Leukotrienes belong to a family of biologically active conjugated trienes that are formed from arachidonic acid via the 5-lipoxygenase pathway and are important mediators of inflammatory reactions. The CysLT1 receptor that specifically serves as receptor for leukotriene D4 (LTD4) has been identified as a G-protein coupled receptor. READ MORE

  4. 4. On the role of gut microbiota in intestinal physiology and hepatic metabolism

    Author : Mattias Bergentall; Göteborgs universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; microbiota; metabolism;

    Abstract : The gut microbiota, a complex and dynamic community of microbes in the mammalian gut, has coevolved with us for ample time providing mutual benefits. However, mechanistic knowledge of these has been limited, but are now becoming increasingly clear. READ MORE

  5. 5. Drug Transport and Metabolism in Rat and Human Intestine

    Author : Sofia Berggren; Hans Lennernäs; Janet Hoogstraate; Patrick Augustijns; Uppsala universitet; []
    Keywords : Biopharmacy; Ussing chamber; microsome; single-pass perfusion; human; rat; intestine; permeability; efflux; P-glycoprotein; metabolism; cytochrome P450; ropivacaine; lidocaine; bupivacaine; Biofarmaci; PPX;

    Abstract : One of the aims of this thesis was to investigate the involvement of efflux proteins, such as the P-glycoprotein (Pgp), in the drug transport in different regions of the rat and the human intestine. The intestinal extrusion of intracellularly formed CYP3A4 metabolites, including whether this extrusion might be mediated by Pgp, was also studied. READ MORE
