  1. 1. Mechanisms of inflammatory lung injury : Studies in isolated perfused rat lungs

    Author : Anders Ljungman; Östergötlands Läns Landsting; []
    Keywords : MEDICINE; MEDICIN;

    Abstract : Isolated perfused rat lungs (IPRLs) were used to study mechanisms andmediators of lung injury due to (i) ischemia-reperfusion (IR) and (ii) endotoxin. To investigate the role of polymorphonuclear neutrophils (PMNs) in IR injury, PMNs were added to the perfusate of IPRLs subjected to IR. READ MORE

  2. 2. Drug absorption in the lungs : studies in the isolated perfused rat lung model combined with physiologically based biopharmaceutics modelling

    Author : Johanna Eriksson; Hans Lennernäs; Erik Sjögren; Helena Thörn; Ben Forbes; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pulmonary drug absorption; physiologically based biopharmaceutics modeling; pulmonary drug delivery; Biofarmaci; Biopharmaceutics;

    Abstract : Pulmonary delivery of drugs is the preferred route of administration for treatment of local lung diseases like asthma and chronic obstructive pulmonary disease. Recently, there has also been increased interest in systemic delivery of drugs via the lungs to avoid problems with low and/or variable gastrointestinal absorption, and as a needle-free alternative for drugs that cannot be ingested. 3. 3. Pulmonary Drug Absorption : In vitro and in vivo investigations of drug absorption across the lung barrier and its relation to drug physicochemical properties

    Author : Ann Tronde; Gerrit Borchard; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; lung; pulmonary; drug delivery; inhalation; drug transport; pharmacokinetics; absorption; bioavailability; drug metabolism; preclinical; rat; in vivo; isolated and perfused lung; Caco-2; nebulization catheter; aerosol; physicochemical; peptide; efflux; FARMACI; PHARMACY; FARMACI; Biopharmaceutics; biofarmaci;

    Abstract : Although, pulmonary drug delivery is a well established means for targeting of drugs to the lungs for the treatment of respiratory diseases as well as for the systemic delivery of volatile anesthetic agents, drug absorption kinetics in the lung have not been subjected to extensive research. The main objective of this thesis was to investigate drug absorption characteristics of the lung barrier, using the isolated and perfused rat lung model and in vivo pharmacokinetic studies in rats. 4. 4. Acute lung failure induced by tricyclic antidepressants : an experimental evaluation

    Author : Karin Svens; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ARDS; tricyclic antidepressants; isolated lungs; rat; endothelin; endothelial cells; tight junctions;

    Abstract : The tricyclic antidepressant drugs (TCA) have, over the years, been reported to be able to cause adult respiratory distress syndrome (ARDS)/acute lung injury in connection with overdose, and are currently among the most common cause of self-poisoning. Approximately 10% of all TCA overdose patients develop ARDS, and the treatment of these patients, irrespective of the event causing ARDS, is mainly supportive. READ MORE

  5. 5. A novel technology for studying the disposition of drugs and toxicants in the lung : short inhalation exposures of the isolated and perfused rat lung to respirable dry particle aerosols

    Author : Pär Ewing; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Aerosol; inhalation; rat; lung; absorption; drugs; carcinogen; particles;

    Abstract : The disposition of solutes following the deposition of inhaled substances in the lung is a complex process that proceeds along parallel paths; passive diffusion of solutes through the air/blood barrier and local metabolism followed by blood-borne clearance to the circulation. Using the isolated and perfused lung (IPL) the pulmonary disposition of inhaled particles can be studied in great detail without interference from distal organs and systemic exposure. READ MORE
