  1. 1. Anti-epileptic and neuroprotective mechanisms by Topiramate and Levetiracetam evaluated in primary cultures

    Author : Mikael Ängehagen; Göteborgs universitet; []
    Keywords : calcium imaging; phosphorylation; kainate; glutamate; caffeine; cerebral cortex; hippocampus; field potentials; neurons; astrocytes;

    Abstract : The aim of this thesis was to evaluate plausible intracellular mechanisms of action for antiepileptic drugs, Topiramate (TPM) and Levetiracetam (LEV), and to investigate if TPM possesses neuroprotective properties. The thesis is based on results from calcium imaging, immunohistochemical stainings, viability tests, and Western blot using primary cortical or hippocampal cultures, or hippocampal brain slices. 2. 2. Modulation of AMPA glutamate receptor functions in primary neuronal cultures

    Author : Gvido Cebers; Karolinska Institutet; Karolinska Institutet; []
    Keywords : glutamate; AMPA; cerebellar granule cells; cortical cells; cyclothiazide; AP-1; transcription factors; receptor desensitization; Ca2 ; neurotoxicity; 45Ca2 uptake; apoptosis; ischemia; muscarinic; carbachol;

    Abstract : Primary neuronal cultures were used to investigate the properties of AMPA receptors and functional consequences of their activation, as well as AMPA receptor desensitization and possibilities for its modulation. We established the presence and spontaneous functional activity of AMPA receptors in cerebellar granule and cortical cell cultures. READ MORE

  3. 3. The phencyclidine model of schizophrenia : dysregulation of brain dopamine systems induced by NMDA receptor antagonists

    Author : Jan M Mathé; Karolinska Institutet; Karolinska Institutet; []
    Keywords : phencyclidine; MK-801; CNQX; prazosin; tetrodotoxin; dopamine; neurotensin; ventral tegmental area; nucleus accumbens; medial prefrontal cortex;

    Abstract : Non-competitive NMDA receptor antagonists, such as phencyclidine (PCP) and dizocilpine (MK-801) can even acutely induce a drug induced psychotic state which closely resembles schizophrenia. The midbrain dopamine (DA) neurons, which originate in the ventral tegmental area (VTA), are involved in motivational, attentional and cognitive processes, and have also been profoundly implicated in schizophrenia. 4. 4. Modulation of cellular mechanisms in a spinal locomotor network

    Author : Jesper Tegnér; Karolinska Institutet; Karolinska Institutet; []
    Keywords : Neuronal Network; Calcium Channels; Calcium-DependentPotassium Channels; Neuronal Oscillator; Neuromodulation; GABAg receptors; Lamprey; Locomotion; Spinal Cord; Computer Simulation;

    Abstract : Neuronal networks generate behaviour. Thus, to understand behaviour requires information on how neuronal networks work in terms of their cellular components. The overall aim of this work has been to link processes occurring at an ionic level to the operation of a neural network specialized for undulatory locomotion. READ MORE

  5. 5. The Role of Cyclooxygenase-2 in Models of Epilepsy and Traumatic Brain Injury : Effects of Selective Cyclooxygenase-2 Inhibitors

    Author : Tina Kunz; Tadeusz Wieloch; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical biosciences; Controlled cortical contusion injury; fluid percussion injury; limbic seizures; nuclear factor-kappa B; prostaglandin metabolism; TUNEL staining; Farmaceutisk biovetenskap; Biopharmacy; Biofarmaci; farmaceutisk farmakologi; Pharmaceutical Pharmacology;

    Abstract : Cyclooxygenase-2 (COX-2) catalyses prostaglandin synthesis from arachidonic acid during inflammation. COX-2 is expressed in the normal brain and is induced in neurological disorders. There is evidence that COX-2 is involved in secondary events leading to cell death in the brain. READ MORE
