  1. 1. Asymmetric transformation of ß- and γ-functionalized alcohols : Study of combined ruthenium-catalyzed racemization and enzymatic resolution

    Author : Annika Träff; Jan-Erling Bäckvall; Reko Leino; Stockholms universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; Kinetic resolution; dynamic kinetic resolution; kinetic asymmetric transformation; dynamic kinetic asymmetric transformation; enzyme catalysis; racemization; asymmetric synthesis; chlorohydrin; cyano acetate; amino alcohol; metalloenzyme; bifunctional catalytic system; Organic synthesis; Organisk syntes; Organic Chemistry; organisk kemi;

    Abstract : The major part of this thesis describes the asymmetric synthesis of β- and γ-amino alcohols through the combination of ruthenium catalyzed racemization and enzymatic kinetic resolution. The dynamic kinetic resolution, DKR, protocol for chlorohydrins was improved by employing Bäckvall’s catalyst, which is a base activated racemization catalyst, in combination with Burkholderia cepacia lipase. 2. 2. Ruthenium-Catalyzed Hydrogen Transfer Reactions : Mechanistic Studies and Chemoenzymatic Dynamic Kinetic Resolutions

    Author : Madeleine Warner; Jan-Erling Bäckvall; Andreas Pfaltz; Stockholms universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; Hydrogen transfer; ruthenium catalysis; kinetic resolution; dynamic kinetic resolution; racemization; asymmetric synthesis; Organic Chemistry; organisk kemi;

    Abstract : The main focus of this thesis lies on transition metal-catalyzed hydrogen transfer reactions. In the first part of the thesis, the mechanism for racemization of sec-alcohols with a ruthenium complex, Ru(CO)2Cl(η5-C5Ph5) was studied. READ MORE

  3. 3. Organocatalytic Acylation for the Kinetic Resolution of Secondary Aryl Alcohols : Synthetic Applications and Mechanistic Studies

    Author : Laura Mesas Sánchez; Peter Dinér; Pavel Kočovský; Uppsala universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; kinetic resolution; organocatalysis; secondary alcohols; reaction progress kinetic analysis; Chemistry with specialization in Organic Chemistry; Kemi med inriktning mot organisk kemi;

    Abstract : The research described in this thesis focuses on the catalytic acylative kinetic resolution (KR) of aromatic secondary alcohols, using a planar-chiral 4-(dimethylamino)pyridine (DMAP) organocatalyst.In the first part of this thesis, the substrate scope of the above mentioned process was expanded to aromatic secondary alcohols that contain an extra functional group in the alkyl moiety, such as 1,2-azido alcohols, 2-hydroxy-2-aryl-ethylphosphonates and 2-hydroxy-2-aryl esters. 4. 4. Chemoenzymatic Resolution in Dynamic Systems : Screening, Classification and Asymmetric Synthesis

    Author : Yan Zhang; Olof Ramström; Sabine Flitsch; KTH; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; constitutional dynamic chemistry; dynamic systemic resolution; dynamic kinetic resolution; enzyme catalysis; transesterification; enzyme promiscuity; asymmetric synthesis; pattern recognition; self-inhibition.;

    Abstract : This  thesis  is  divided  into  four  parts,  all  centered  around  Constitutional Dynamic  Chemistry  (CDC)  and  Dynamic  Kinetic  Resolution  (DKR)  using biocatalysts for selective transformations, and their applications in screening of bioactive compounds, organic synthesis, and enzyme classification.   In  part  one,  an  introduction  to  CDC  and  DKR  is  presented,  illustrating  the basic  concepts,  practical  considerations  and  potential  applications  of  such dynamic systems, thus providing the background information for the studies in the following chapters. 5. 5. Enantioselective Synthesis of Sec-Alcohol Derivatives and Diols via Combined Ruthenium and Enzyme Catalysis

    Author : Linnéa Borén; Jan-Erling Bäckvall; Romas Kazlauskas; Stockholms universitet; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; catalysis; kinetic resolution; ruthenium catalyst; Dynamic kinetic resolution; Dynamic Kinetic Asymmetric Transformation; Organic chemistry; Organisk kemi; Organic Chemistry; organisk kemi;

    Abstract : The first part of this thesis describes the synthesis of enantiopure secondary alcohol derivatives. These syntheses are carried out via the combination of an enzyme as a resolution catalyst and a ruthenium catalyst as a racemization catalyst, in what is called dynamic kinetic resolution (DKR).
