  1. 6. Direct Thrombin Inhibitors in Treatment and Prevention of Venous Thromboembolism: Dose – Concentration – Response Relationships

    Author : Marie Cullberg; Mats O. Karlsson; Ulf G. Eriksson; Nick H.G. Holford; Uppsala universitet; []
    Keywords : Pharmaceutical biosciences; Ximelagatran; pharmacokinetic; pharmacodynamic; activated partial thromboplastin time; utility function; dosing strategy; venous thromboembolism; NONMEM; Farmaceutisk biovetenskap;

    Abstract : For prevention and treatment of thrombotic diseases with an anticoagulant drug it is important that an adequate dose is given to avoid occurrence or recurrence of thrombosis, without increasing the risk of bleeding and other adverse events to unacceptable levels. The aim of this thesis was to develop mathematical models that describe the dose-concentration (pharmacokinetic) and concentration-response (pharmacodynamic) relationships of direct thrombin inhibitors, in order to estimate optimal dosages for treatment and long-term secondary prevention of venous thromboembolism (VTE). 2. 7. Nasal Drug Delivery : In Vitro Studies on Factors Influencing Permeability and Implications on Absorption

    Author : Cecilia Wadell; Frans Merkus; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacy; FARMACI; PHARMACY; FARMACI; galenisk farmaci; Pharmaceutics;

    Abstract : Nasal delivery is a feasible alternative to oral or parenteral administration for some drugs because of the high permeability of the nasal epithelium, rapid drug absorption across this membrane and avoidance of hepatic first-pass metabolism. The main objective of this thesis was to investigate factors influencing the permeability of the nasal mucosa to various compounds and to evaluate implications for drug absorption via the nasal route. 3. 8. Experimental studies on thrombosis and thrombolysis : With special reference to importance of lys-plasminogen, active site thrombin inhibitors and stable fish oil

    Author : Liying Chen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Surgery; Thrombolysis; thrombosis; recombinant tissue-type plasminogen activator; lys- and glu-plasminogen; aspirin; thrombin inhibitors; platelets; superoxide dismutase; stable fish oil; Kirurgi; Surgery; Kirurgi; rättsmedicin; Forensic Medicine;

    Abstract : Dissolution and prevention of thrombus in the atherosclerotic coronary artery have become an important part of treatment therapy of acute myocardial infarction. However, the currently most effective thrombolytic agent, recombinant tissue-type plasminogen activator (rt-PA), only elicits 80% recanalization of thrombotically occluded coronary arteries and early occlusion occurs in about 40% of successfullyrecanalized vessels. 4. 9. Endogenous t-PA release and pharmacological thrombolysis - Experimental animal studies of the coronary circulation

    Author : Jan-Arne Björkman; Göteborgs universitet; []
    Keywords : Klinisk fysiologi;

    Abstract : Abstract The physiologically most important activator of intravascular fibrinolysis is tissue-type plasminogen activator (t-PA) released from endothelial cells. In man, sympathomimetic drugs increase the systemic concentration of t-PA. READ MORE

  5. 10. Human liver in vitro models for evaluation of drug metabolism and disposition

    Author : Malin Darnell; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The administrated dose of a drug is adjusted to give a therapeutic effect in patients without causing side-effects or toxicity. Cytochrome P450 (P450) and UDP-glucuronosyltransferase (UGT) enzymes, uptake and efflux transporters and nuclear receptors regulating these enzymes, expressed in the liver and in other tissues, are all important players in drug metabolism, disposition and elimination.
