  1. 1. Antimicrobial Peptide Interactions with Phospholipid Membranes : Effects of Peptide and Lipid Composition on Membrane Adsorption and Disruption

    Author : Adam A Strömstedt; Martin Malmsten; Georges Belfort; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; antimicrobial; peptide; adsorption; liposome; membrane; melittin; cholesterol; tryptophan; phospholipid; bilayer; PHARMACY; FARMACI; Pharmaceutical Physical Chemistry; farmaceutisk fysikalisk kemi;

    Abstract : The interactions between antimicrobial peptides and phospholipid membranes were investigated, in terms of lipid headgroup variations and the role of cholesterol, as well as peptide composition and structure. Also strategies for increasing proteolytic stability were evaluated. READ MORE

  2. 2. Nanosized Bilayer Disks as Model Membranes for Interaction Studies

    Author : Anna Lundquist; Katarina Edwards; Marja-Liisa Riekkola; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Disk; disc; lipid bilayer; PEG-lipid; model membrane; interaction; partitioning; drug; liposome; cryo-TEM; neutron scattering; capillary electrophoresis; immobilization; biosensor; membrane protein; melittin; Chemistry; Kemi;

    Abstract : PEG-lipid stabilized bilayer disks have been found in lipid mixtures containing polyethylene glycol (PEG)-lipids where the combination of a high bending rigidity and low PEG-lipid/lipid miscibility favours disk formation. The disks are planar and circular in shape and their long-term stability is excellent. READ MORE

  3. 3. Physico-Chemical Investigations of, and Characterization of Model Membranes for, Lipid-Peptide Interactions

    Author : Per Wessman; Katarina Edwards; Martin Malmsten; Michel Lafleur; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Physical chemistry; Fysikalisk kemi; Physical Chemistry; fysikalisk kemi;

    Abstract : The main focus of this thesis is on the effects caused by α-helical peptides on liposome structure, the impact of cholesterol on the peptide-membrane interactions, and the effect of aggregate curvature on the peptide affinity. Results of the studies show that the membrane destabilizing effect of the cationic α-helical peptide melittin is modulated by cholesterol. READ MORE

  4. 4. Development of lipodisks as carriers for cationic amphiphilic peptides

    Author : Karin Reijmar Brunnström; Katarina Edwards; Fredrik Höök; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES;

    Abstract : Antibiotics have made a tremendous contribution to mankind. They are one of the most successful medicines in human history. However, more and more bacterial strains develop resistance and the risk to public health can hardly be overstated. New types of antibiotics are urgently needed. READ MORE

  5. 5. Development and Evaluation of Lipodisks Intended for Use as Biomimetic Membranes and Drug Carriers

    Author : Malin Morin Zetterberg; Katarina Edwards; Jonas Bergquist; Ana Maria Carmona-Ribeiro; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; model membranes; drug delivery; drug partitioning; antimicrobial peptides; nanocarriers; cryo-TEM; polymer-stabilized bilayer disks; Chemistry with specialization in Physical Chemistry; Kemi med inriktning mot fysikalisk kemi;

    Abstract : Polyethylene glycol-stabilized lipodisks have emerged as a novel type of lipid-based nanoparticles with high potential as both drug carriers and biomimetic membranes. In this thesis we assess both of these applications, and show how the properties of the lipodisks can be further developed and optimized. READ MORE
