  1. 16. Targeting double-stranded DNA with locked nucleic acid-based (LNA) oligonucleotides (ON) and DNA-binding peptide conjugates

    Author : Y Vladimir Pabón-Martínez; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : In this thesis we have studied two different types of molecules that bind to the major groove of the DNA: (i) oligonucleotides (ONs) containing locked nucleic acid (LNA) and in combination with intercalating agents (Twisted Intercalating Nucleic Acid, TINA, and Benzoquinoquinoxaline, BQQ) and (ii) peptides mimicking the leucine zipper transcription factor (GCN4 bZIP TF). Both ONs and peptides bind to the DNA double helix aiming to serve as transcription regulators of disease related genes. READ MORE

  2. 17. Studies on Transformations of H-Phosphonates into DNA Analogues Containing P-S or P-C Bonds

    Author : Tommy Johansson; Jacek Stawinski; Joachim Wolfgang Engels; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Organisk kemi; Organic chemistry; Organisk kemi;

    Abstract : In this thesis, mechanistic and synthetic studies on transformations of H-phosphonates into DNA analogues containing P-S or P-C bonds are described. Configurational stability of dinucleoside H-phosphonates and the stereochemical course of their sulfurisation in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) were investigated. READ MORE

  3. 18. Rational modifications of cell-penetrating peptides for drug delivery : Applications in tumor targeting and oligonucleotide delivery

    Author : Maarja Mäe; Ülo Langel; Edvard Smith; Stockholms universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell-penetrating peptide; oligonucleotide; drug delivery; tumor targeting; Neurochemistry; Neurokemi; Neurochemistry and Molecular Neurobiology; neurokemi med molekylär neurobiologi;

    Abstract : High molecular weight biomolecules are becoming important in the development of new therapeutics. However, their size and nature creates a major limitation for their application – poor penetration through biological membranes. READ MORE

  4. 19. Design and application of RNA therapeutics for splice site mutations

    Author : Burcu Bestas; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Precursor messenger RNA splicing is one of the most fundamental and complex mechanisms in eukaryotes. Over 90% of the human genes undergo alternative splicing, which is essential for the regulation of gene expression. A dynamic RNA-protein complex called the spliceosome catalyzes splicing. READ MORE

  5. 20. Cell-penetrating peptides; chemical modification, mechanism of uptake and formulation development

    Author : Kariem Ezzat; Ülo Langel; Steven Dowdy; Stockholms universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cell penetrating peptides; gene therapy; scavenger receptors; pharmaceutical formulation; solid dispersion; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Abstract : Gene therapy holds the promise of revolutionizing the way we treat diseases. By using recombinant DNA and oligonucleotides (ONs), gene functions can be restored, altered or silenced according to the therapeutic need. READ MORE
