21. On the evolutionary effects of linkage and deleterious mutations in small populations
Abstract : The aim of this thesis is to evaluate the effect of deleterious mutations and linkage on variation in neutral markers and fitness in small populations. The study is based on two main approaches, one by using computer simulations, the other by exploring genetic variation in a cyclical parthenogen, Daphnia pulex (Crustacea: Cladocera). READ MORE

22. Surviving the ratchet : Modelling deleterious mutations in asexual populations
Abstract : One of the most unforgiving processes in nature is that of Muller's ratchet, a seemingly irreversible accumulation of deleterious mutations that all organisms have to deal with or face extinction. The most obvious way to avoid fitness collapse is recombination, though asexual populations usually do not have the luxury of recombining freely. READ MORE

23. Mechanisms of adaptive mutations in bacteria
Abstract : Bacteria have the ability to adapt to various challenging environments. The rate, extent and consequences of bacterial adaptation depend on both microbial and environmental factors. READ MORE

24. Alzheimer's disease mutations and cellular signalling
Abstract : Alzheimer's disease is the most frequent cause of dementia in elderly and is characterised neuropathologically by the extracellular deposition of amyloid plaques (containing the 39-43 amino acid Abeta peptide), as well as intracellular neurofibrillary tangles (NFTs) composed primarily of an abnormally hyperphosphorylated form of the microtubule-associated protein tau. A proportion (10- 15%) of Alzheimer's disease cases is familial, the remainder being sporadic. READ MORE

25. Understanding Cancer Mutations by Genome Editing
Abstract : Mutational analyses of cancer genomes have identified novel candidate cancer genes with hitherto unknown function in cancer. To enable phenotyping of mutations in such genes, we have developed a scalable technology for gene knock-in and knock-out in human somatic cells based on recombination-mediated construct generation and a computational tool to design gene targeting constructs.
