  1. 1. Regulation of inducible nitric oxide synthase - Consequences in experimental models of bladder disease

    Author : Rebecka Johansson; Avdelningen för klinisk kemi och farmakologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacy; toxicology; Farmakologi; smooth muscle-myosin heavy chain; Pharmacological sciences; pharmacognosy; farmakognosi; farmaci; actin; BDNF; Rho; aminoguanidine; TNF-alpha; IL-1beta; hypertrophy; differentiation; smooth muscle; bladder; nitric oxide; inducible nitric oxide synthase; toxikologi;

    Abstract : Inducible nitric oxide synthase (iNOS) expression is associated with various pathophysiological conditions in the lower urinary tract. The aims of this thesis have been to investigate the regulation of iNOS in bladder smooth muscle cells and the consequences on neuromuscular regulation, cell growth and differentiation. READ MORE

  2. 2. Islet constitutive nitric oxide synthase and nitric oxide production. Modulatory effects on insulin and glucagon secretion

    Author : Björn Åkesson; Allmänmedicin och klinisk epidemiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacognosy; Pharmacological sciences; mouse; carbachol; muscarinic agonist; cyclic nucleotides; calcium-fluxes; NOS-inhibitors; in vitro and in vivo; insulin and glucagon secretion; Nitric oxide NO ; constitutive nitric oxide synthase cNOS ; pharmacy; toxicology; Farmakologi; farmakognosi; farmaci; toxikologi; Endocrinology; secreting systems; diabetology; Endokrinologi; sekretion; diabetologi;

    Abstract : The aim of this thesis was to elucidate the role of nitric oxide (NO) in the transduction signalling of insulin and glucagon release by stimulating or inhibiting islet endogenous NO-production as well as by using NO donors. Insulin release stimulated by L-arginine, the substrate for constitutive NO-synthase (cNOS)-catalyzed NO-production, was increased by the selective NOS-inhibitor NG-nitro-L-arginine methyl ester (L-NAME) and suppressed by the intracellular NO donor hydroxylamine. 3. 3. Regulation of cytokine-induced nitric oxide production in insulin-producing cells

    Author : Malin Flodström; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell biology; Diabetes; pancreatic islets; insulin-producing cells; cytokines; nitric oxide; inducible nitric oxide synthase; nuclear factor-kappaB; interferon regulatory factor-1; arginine; citrulline; argininosuccinate synthetase; insulin release; Cellbiologi; Cell biology; Cellbiologi; medicinsk cellbiologi; Medical Cell Biology;

    Abstract : Cytokine-induced expression of the inducible form of nitric oxide synthase (iNOS) and production of nitric oxide (NO) may contribute to pancreatic β-cell damage during the development of type 1 diabetes. iNOS catalyses the conversion of arginine into citrulline and NO, a reaction regulated by the amount of expressed iNOS enzyme and by the cellular availability of arginine. 4. 4. Regulation of Insulin Secretion in Relation to Nitric Oxide, Carbon Monoxide and Acid alpha-Glucoside Hydrolase Activities

    Author : Henrik Mosén; Lund Klinisk fysiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Endokrinologi; secreting systems; diabetology; Endocrinology; Medicin människa och djur ; Medicine human and vertebrates ; GK rat; insulin secretion; carbon monoxide; acid alpha-glucoside hydrolases; nitric oxide; heme oxygenase; isoforms of nitric oxide synthase; diabetologi; sekretion;

    Abstract : The main stimulus for insulin secretion is elevated blood glucose levels. In this thesis the aim was to study less well-known systems involved in the complex regulation of glucose-stimulated insulin release, emphasizing the role of the lysosomal/vacuolar system and the associated glycogenolytic acid alpha-glucoside hydrolases, but also the HO-CO and NOS-NO systems. 5. 5. Post-Transcriptional Regulation of the Murine Inducible Nitric Oxide Synthase Gene

    Author : Malin Söderberg; Matti A. Lang; Françoise Raffalli-Mathieu; Eeva Moilanen; Uppsala universitet; []

    Abstract : Large amounts of nitric oxide (NO) are produced by the inducible nitric oxide synthase (iNOS) upon inflammatory stimuli. NO is a multifaceted molecule, which may have beneficial effects as an antimicrobial agent in the immune defense, or cytotoxic effects in chronic inflammations, manifested as e.g. arthritis and asthma.
