1. Conformationally Constrained Oligonucleotides for RNA Targeting
Abstract : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. READ MORE

2. Targeting RNA by the Antisense Approach and a Close Look at RNA Cleavage Reaction
Abstract : This thesis summarizes the results of studies on two aspects of nucleic acids. Chemically modified antisense oligonucleotides (AONs) have been evaluated with regards to their suitability for mRNA targeting in an antisense approach (Paper I – III). READ MORE

3. Development and synthesis of oligonucleotide building blocks and conjugates for therapeutic applications
Abstract : The research and development of oligonucleotide therapeutics has been a topic of great importance in life sciences. The potential to treat rare genetic diseases can be demonstrated simply by highlighting the increasing amount of approved oligonucleotide-based drugs. READ MORE

4. Biosensing platforms using graphene based bioreactive nanostructures with various dimensions
Abstract : Nanomaterials have brought new aspects and improvements to the biosensing field due to their unique physical and chemical properties that are not shown in the bulk state. This thesis focuses on concepting, developing and testing of biosensors where nanomaterials including graphene gold nanoparticles (AuNPs) and magnetic nanoparticles (MNPs) constitute the biosensors. 5. Epigenetic Control of Mammalian Development : Studies on an imprinting control region
Abstract : A subset of autosomal genes is preferentially or exclusively expressed from one of the parental alleles. This phenomenon, termed genomic imprinting, is highlighted by the neighboring Igf2 and H19 genes, which are monoallelically expressed on opposite parental chromosomes. These features are governed by a 2. READ MORE
