17. Targeting double-stranded DNA with locked nucleic acid-based (LNA) oligonucleotides (ON) and DNA-binding peptide conjugates
Abstract : In this thesis we have studied two different types of molecules that bind to the major groove of the DNA: (i) oligonucleotides (ONs) containing locked nucleic acid (LNA) and in combination with intercalating agents (Twisted Intercalating Nucleic Acid, TINA, and Benzoquinoquinoxaline, BQQ) and (ii) peptides mimicking the leucine zipper transcription factor (GCN4 bZIP TF). Both ONs and peptides bind to the DNA double helix aiming to serve as transcription regulators of disease related genes. READ MORE

Abstract : This thesis is based on nine research publications (I – IX) on structure and reactivity of RNA vis-à-vis DNA. The DNA and RNA are made of flexible pentose sugar units, polyelectrolytic phosphodiester backbone, and heterocyclic nucleobases. Abstract : The WT1 protein, a product of the Wilms’ tumour 1 gene (WT1) is a zinc finger transcription factor implicated in a number of cellular processes particularly involved in the development of the urogenital system. Mutations in this gene have been implicated in abnormal development of the urogenital system resulting in syndromes such as the Denys-Drash, WAGR and Frasier and in the childhood kidney malignancy, Wilms’ tumour.
