1. Retroviral and related nucleic acid processing enzymes expression and detection : Applications on clinical studies of HIV and HERVs
Abstract : Reverse transcriptase (RT), the expression and detection of the enzyme marking the family Retroviridae, has been studied. A new RT activity assay adapted to 96-well microtitre plates is described. It uses a non- radioactive bromodeoxyuridine triphosphate (BrdUTP) substrate in a solid phase format. The assay was highly sensitive. READ MORE

2. Development of novel DNA vaccine and immunotherapeutic approaches
Abstract : Many potential DNA vaccination and immunotherapeutic strategies has been explored in both animal models and human settings in recent years. However, the failure of efficient and safe method development suggests that it is time to rethink, since more innovative approaches are needed for better control of infection and immunotherapy. 3. Alphavirus vectors as recombinant vaccines
Abstract : This thesis describes further developments of an expression system based on the alphavirus Semliki Forest virus (SFV), and its potential use as recombinant vaccine. The RNA genome of SFV contains a 3'open reading frame (ORF) encoding the structural proteins, and a 5' ORF coding for the viral replicase. READ MORE

4. Viral vectors, in innate and adaptive immunity
Abstract : Recombinant viral vectors and DNA vectors induce strong immune responses in animal models. However, in clinical trials, the generation of immune responses is less robust, suggesting that further optimization and a deeper understanding of nucleic acid-based vaccines are required. READ MORE

5. Production and delivery of recombinant subunit vaccines
Abstract : Recombinant strategies are today dominating in thedevelopment of modern subunit vaccines. This thesis describesstrategies for the production and recovery of protein subunitimmunogens, and how genetic design of the expression vectorscan be used to adapt the immunogens for incorporation intoadjuvant systems.
