  1. 1. Energy-Related Metabolites during and after Induced Myocardial Ischemia and Retroperfusion of the Coronary Sinus : An Experimental Study in the Pig with the Microdialysis Technique and Extracorporeal Circulation

    Author : Vitas Zemgulis; Rolf Ekroth; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Surgery; Ischemia; myocardial infarction; reperfusion; energy metabolism; coronary sinus retroperfusion; microdialysis; taurine; nucleoside transport; adenosine; aerobic lactate production; Kirurgi; Surgery; Kirurgi; Thoracic and Cardivascular Suregery; kirurgi; särskilt thoraxkirurgi;

    Abstract : In the clinical setting great efforts have been made with contradictory results to operate upon acutely myocardial ischemic patients. The reasons for the absence of unambiguous results are not well understood nor are they scientifically explored. READ MORE

  2. 2. Distribution of antiviral nucleoside analogues into brain and skin

    Author : Natalia Borg; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The aim of this thesis was to study physiological and physico-chemical factors influencing the pharmacokinetics and distribution over the blood-brain barrier of antiviral nucleoside analogues in the rat, and to study the distribution to the human skin in vivo of an anti-herpetic nucleoside analogue. Microdialysis was used to sample the free (unbound) extracellular concentration of the nucleoside analogues from the brain tissue, muscle and blood in the rat and from the dermis of healthy human volunteers. 3. 3. Pharmacological and molecular investigations on the mechanisms underlying resistance of human leukaemia cells to the antimetabolites methotrexate, 6-mercaptopurine and 6-thioguanine

    Author : Alan Kambiz Fotoohi; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The goal of this thesis has been to elucidate the mechanisms underlying resistance to methotrexate (MTX), 6-mercaptopurine (6-MP) and 6-thioguanine (6-TG), the antimetabolites widely used in treatment of childhood leukemia, as well as to determine the influence of 7-hydroxymethotrexate (7-OHMTX), the major metabolite of MTX, on the therapeutic action of MTX. Resistant sublines of leukemic cell lines were developed by long-term exposure to stepwise increasing concentrations of these different agents. READ MORE

  4. 4. Crystallographic studies of gluconate kinase

    Author : Louise Kraft; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : The first steps of gluconate metabolism in Escherichia coli are the transport of gluconate into the cell by a gluconate transport system, followed by phosphorylation to gluconate-6- phosphate by gluconate kinase. There exist two such different pathways, Grill and GntIl, respectively. READ MORE

  5. 5. Pharmacological studies on uptake, metabolism, and resistance to anti-cancer drugs : insights into the treatment of leukemia

    Author : Hazhar Karim; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Leukemia is blood cancer that begins in the bone marrow and characterized by abnormal production of white blood cells. Due to advances in treatment, there has been a dramatic increase in survival rate for patients with different types of leukemia. READ MORE
