  1. 21. Development of DNA-based methods for analysis of protein interactions

    Author : Axel Klaesson; Ola Söderberg; Mats Nilsson; Cristina Al-Khalili Szigyarto; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; In situ proximity ligation assay PLA ; rolling circle amplification RCA ; Hybridization chain reaction HCR ; proxHCR; Oligonucleotide design; Protein-protein interactions; Post-translational modifications.; Farmaceutisk vetenskap; Pharmaceutical Science; Molekylär cellbiologi; Molecular Cellbiology;

    Abstract : In situ proximity ligation assay (PLA) is a method for detection of protein interactions, post-translational modifications (PTMs) and individual proteins that allows information about their localization in a cell or tissue to be extracted. The method is based on oligonucleotide-conjugated antibodies (proximity probes) that upon binding of two epitopes in close proximity give rise to an amplifiable DNA circle. READ MORE

  2. 22. Advancing DNA-based proximity methods

    Author : Mattias Leino; Ola Söderberg; John Löfblom; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; in situ; proximity dependent intiation of hybridization chain reaction; hybridization chain reaction; Oligonucleotide design; proximity ligation assay; protein interaction; microscopy; purification; Biology with specialization in Molecular Biotechnology; Biologi med inriktning mot molekylär bioteknik;

    Abstract : Cellular functions are governed by intricate chains of interactions between proteins. In order to properly understand cellular biology one must look into, not only protein function, but also its interacting network. READ MORE

  3. 23. Conformationally Constrained Oligonucleotides for RNA Targeting

    Author : Qing Li; Jyoti Chattopadhyaya; Li-He Zhang; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; conformationally constrained nucleoside; antisense oligonucleotide; RNA affinity; nuclease stability; RNase H; RNA degradation; Chemistry with specialization in Bioorganic Chemistry; Kemi med inriktning mot bioorganisk kemi;

    Abstract : A short oligonucleotide sequence as in a single-stranded antisense oligo nucleotides (AON) or in double-stranded small interfering RNAs (siRNA) can modulate the gene expression by targeting against the cellular mRNA, which can be potentially exploited for therapeutic purposes in the treatment of different diseases. In order to improve the efficacy of oligonucleotide-based drugs, the problem of target affinity, nuclease stability and delivery needs to be addressed. READ MORE

  4. 24. Characterization of parameters influencing intracellular bioavailability and prediction of intracellular drug exposure

    Author : Andrea Treyer; Per Artursson; Gert Fricker; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intracellular drug bioavailability; unbound drug concentration; drug disposition; ADME; drug transport; drug metabolism membrane partitioning; phospholipid; drug-drug interaction; antisense oligonucleotide; cell-penetrating peptide; Farmaceutisk vetenskap; Pharmaceutical Science;

    Abstract : This thesis work investigates factors influencing intracellular drug disposition. An experimental method for measurement of intracellular bioavailability (Fic), was used throughout. Fic is defined as the ratio between the unbound drug concentration inside the cell and the compound concentration in the cell exterior. READ MORE

  5. 25. Cell-Penetrating Peptides for Mitochondrial Targeting

    Author : Carmine Pasquale Cerrato; Ülo Langel; Shana O. Kelley; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Mitochondrial targeting; cell-penetrating peptides; antioxidant activity; scavenging ability; oligonucleotide delivery; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Abstract : Mitochondria have simply been known as the cell’s powerhouse for a long time, with its vital function of producing ATP. However, substantially more attention was directed towards these organelles once they were recognized to perform several essential functions having an impact in cell biology, pharmaceutics and medicine.
