    Author : Marko Filipović; Jeanette Hellgren Kotaleski; Arvind Kumar; Gilad Silberberg; Ad Aertsen; Andrew Sharott; KTH; []
    Keywords : NATURAL SCIENCES; NATURVETENSKAP; NATURVETENSKAP; NATURAL SCIENCES; basal ganglia; striatum; medium spiny neurons; subthalamic nucleus; globus pallidus; Parkinson s disease; in vivo; oscillations; spectral analysis; response variability; networks; simulations; modelling; Numerical Analysis; Numerisk analys; Computer Science; Datalogi; Biological and Biomedical Physics; Biologisk och biomedicinsk fysik;

    Abstract : Striatal medium spiny neurons (MSNs) play a crucial role in various motor and cognitive functions. They are separated into those belonging to the direct pathway (dMSNs) and the indirect pathway (iMSNs) of the basal ganglia, depending on whether they express D1 or D2 type dopamine receptors, respectively. 2. 2. PET and the Multitracer Concept: An Approach to Neuroimaging Pathology

    Author : Henry Engler; Bengt Långström; Göran Hedenstierna; Juha Rinne; Uppsala universitet; []
    Keywords : Neurosciences; PET Positron Emission Tomography ; multitracer; neuroimaging pathology; astrocytes; microglia; neurodegeneration; amyloid; AD; CJD; PD; Neurovetenskap;

    Abstract : Patients suffering from different forms of neurodegenerative diseases, such as: Creutzfeldt Jacob Disease (CJD), Alzheimer disease (AD), mild cognitive impairment (MCI), frontotemporal dementia and Parkinson’s disease (PD) were examined with Positron Emission Tomography (PET) and the combination of different radiotracers: 15O-water, N-[11C-methyl]-L-deuterodeprenyl (DED), [18F] 2-fluorodeoxyglucose: (FDG), N-methyl-[11C]2-(4-methylaminophenyl)-6-hydroxybenzothiazole (PIB) and L-[11C]-3,4-dihydroxiphenyl-alanine (DOPA). The radiotracers and the combinations of different radiotracers were selected with the intention to detect, in the brain, patterns of neuronal dysfunction, astrocytosis, axon degeneration or protein aggregation (amyloid), in the brain which are pathognomonic for specific diseases and may contribute to improve clinical differential diagnoses.     Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICAL AND HEALTH SCIENCES; Cerebrospinal fluid CSF ; biomarkers; therapeutic targets; neurodegeneration; Alzheimer’s disease AD ; Parkinson’s disease PD ; Corticobasal degeneration CBD ; Progressive supraneuclear palsy PSP ; Lysosomes; Endosomes; Autophagy; LAMP-2; Lysozyme;

    Abstract : The pre-symptomatic stage of neurodegenerative diseases such as Alzheimer’s disease (AD) and Parkinson’s disease (PD) occurs several decades before the clinical onset. Changes in the lysosomal network, i.e. the autophagosomal, endosomal and lysosomal vesicular system, are among the first alterations observed. READ MORE

  4. 4. Islet Amyloid Polypeptide: Interaction with Amyloid Beta, Alpha-Synuclein and BRICHOS

    Author : Ye Wang; Gunilla Westermark; Ludmilla Morozova-Roche; Uppsala universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; amyloid; IAPP; Abeta; alpha-synuclein; Bri2 BRICHOS; luminescent conjugated oligothiophenes; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract :  Amyloid, the congophilic deposits of misfolded protein, are pathological hallmarks of many common diseases, among others, Alzheimer’s disease (AD), Parkinson’s disease (PD), and type 2 diabetes (T2D). Amyloid beta (Aβ) forms senile plaques in AD, alpha-synuclein (aSyn) forms Lewy bodies and Lewy neurites in PD, islet amyloid polypeptide (IAPP) forms islet amyloid in T2D. READ MORE

  5. 5. Apolipoprotein E and alpha-synuclein interactions, and their associations with neurodegenerative diseases

    Author : Daniel Twohig; Henrietta M. Nielsen; Andreas Barth; Tiago Fleming Outeiro; Stockholms universitet; []
    Keywords : MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neurokemi med molekylär neurobiologi; Neurochemistry with Molecular Neurobiology;

    Abstract : The apolipoprotein E gene (APOE) is expressed as three major isoforms, APOE ε2, ε3 and ε4 that encode the apoE2, E3 and E4 proteins, respectively. APOE ε4 is associated with an increased risk for age-related neurodegenerative diseases including Alzheimer’s disease (AD) and dementia with Lewy bodies (DLB), and increases the risk of dementia in Parkinson’s disease (PD), however, the exact molecular mechanisms underpinning the associations between APOE ε4 and neurodegeneration are still unknown.
