  1. 1. An Investigation of Peroxidase Activity in Biomimetic and Biological Systems

    Author : Ekaterina Ryabova; Kemiska institutionen; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; metabolism; Metabolism; enzymologi; Proteiner; enzymology; Proteins; hemoglobin; catalysis; peptide; peroxidase; protoporphyrin IX; Biokemi; Biochemistry;

    Abstract : In order to create a heme environment which permits biomimicry of heme-containing plant peroxidases and investigate the role played by the vinyl side chains of the porphyrin ring on the stability and, possibly, catalytic activity of model compounds, a number of new synthetic microperoxidases -– hemin-6(7)-gly-his methyl ester (HGH) and hemin-6(7)-gly-gly-his methyl ester (HGGH) and hemin-6,7-bis(gly-gly-his methyl ester) (H2GGH) as well as mesohemin-6(7)-gly-gly-his methyl ester (MGGH) and deuterohemin-6(7)-gly-gly-his methyl ester (DGGH) have been prepared by condensation of a peptide residue with the propionic side chains of hemin, mesohemin and deuterohemin, respectively. Reactivity studies towards different organic and inorganic substrates showed that not only five- but also six-coordinate hemin peptide complexes catalyze the oxidation of substrates by H2O2. 2. 2. Peptide-tagged proteins in aqueous two-phase systems

    Author : Anna Nilsson; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Metabolism; Biokemi; metabolism; Biochemistry; green fluorescent protein; cutinase; tryptophan; Aqueous two-phase system; peptide-tagged protein;

    Abstract : This thesis deals with proteins containing peptide tags for improved partitioning in aqueous two-phase systems. Qualitatively the peptide-tagged protein partitioning could be predicted from peptide data, i.e. partitioning trends found for peptides were also found for the peptide-tagged proteins. READ MORE

  3. 3. Combinatorial Methodology. Screening for affinity specific ligands to biological and artificial receptors

    Author : Margareta Krook; Tillämpad biokemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; peptide library; combinatorial; screening; phage display; affinity; DNA-interaction; chymotrypsin; peptide inhibitor; antibody; IgG; Fc; molecular imprinting; steroid; receptor; Biochemistry; Metabolism; Biokemi; Biotechnology; Bioteknik;

    Abstract : Combinatorial screening methodology has been employed to select new affinity specific ligands to bilogical and artificial receptors. The screening methodology, in particular screening of combinatorial phage display peptide libraries against target molecules differing in function and shape, is focused upon. READ MORE

  4. 4. Protein Surfaces Probed by Aqueous Two-phase Partitioning

    Author : Kristina Berggren; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Metabolism; partitioning; Aqueous two-phase systems; protein surface; peptide; Biochemistry; amino acid residue; Biokemi;

    Abstract : This thesis is concerned primarily with peptide and protein partitioning in aqueous two-phase systems and the connection between partitioning behavior and protein structure. The surface properties of a protein is often crucial for recognition and interaction with other molecules. READ MORE

  5. 5. The conserved methionines in the chloroplast small heat shock protein - Role in chaperone-substrate interactions and effects of methionine sulfoxidation on chaperone activity

    Author : Niklas Gustavsson; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Metabolism; Biochemistry; oxidative stress; thermostability; temperature stress; stress tolerance; sHsp; protein aggregation; molecular chaperone; Heat stress; methionine sulfoxidation; Biokemi; metabolism; Plant biochemistry; Växtbiokemi;

    Abstract : The chloroplast-localized small heat shock protein (sHsp) Hsp21 belongs to the family of alpha-crystallin like sHsps which form large oligomeric structures and protect partly unfolded aggregation-prone proteins against aggregation. The main focus for this thesis has been on the highly conserved methionines in Hsp21. READ MORE
