  1. 1. On the Generation of cAMP Oscillations and Regulation of the Ca2+ Store-operated Pathway in Pancreatic Islet α- and β-cells

    Author : Geng Tian; Erik Gylfe; Anders Tengholm; Patrick Gilon; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cAMP; oscillations; adrenaline; GLP-1; phosphodiesterase; palmitate; STIM1; Orai1; store-operated calcium entry; insulin secretion; glucagon secretion; β-cell; α-cell; Medical Cell Biology; Medicinsk cellbiologi;

    Abstract : Insulin and glucagon are released in pulses from pancreatic β- and α-cells, respectively. Both cell types are electrically excitable, and elevation of the cytoplasmic Ca2+ concentration ([Ca2+]i) due to depolarization with voltage-dependent entry of the cation is the main trigger of hormone secretion. 2. 2. Cyclic Nucleotide Phosphodiesterase 3B: Regulation in rat adipocytes and 3T3-L1 cells

    Author : Tova Landström; Institutionen för experimentell medicinsk vetenskap; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; 3T3-L1 TNF-alpha; adipocyte; insulin; p70S6 kinase; mitogen-activated protein kinase; protein kinase B; phosphatidylinositol-3 kinase; phosphodiesterase 3B; cAMP; lipolysis; phosphorylation; signal transduction; Biochemistry; Metabolism; Biokemi; metabolism;

    Abstract : Insulin stimulation of rat adipocytes results in phosphorylation and activation of the cyclic nucleotide phosphodiesterase 3B (PDE3B), a key enzyme in the antilipolytic signalling pathway of this hormone. In this thesis, the site phosphorylated in PDE3B upon stimulation of rat adipocytes with insulin and/or the beta-adrenergic agonist, isoproterenol is identified. READ MORE

  3. 3. Genetic aspects of stroke : association and linkage studies in a northern Swedish population

    Author : Per-Gunnar Wiklund; Birgitta Stegmayr; Kjell Asplund; Dan Holmberg; Alun Evans; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Internal medicine; stroke; genetics; polymorphism; association; linkage; risk factors; plasminogen activator inhibitor-1; tissue plasminogen activator; angiotensin converting enzyme; angiotensin II receptor type 1; phosphodiesterase 4D; Invärtesmedicin; Internal medicine; Invärtesmedicin; medicin; Medicine;

    Abstract : Stroke is a common, multifactorial cardiovascular disease. A stroke event is the result of traditional risk factors (i.e. hypertension, diabetes, smoking), environmental exposures and genetic factors in a complex interplay. READ MORE

  4. 4. Hypothyroidism and Pregnancy

    Author : Michaela Granfors; Anna-Karin Wikström; Inger Sundström-Poromaa; Helena Åkerud; Ulla Feldt-Rasmussen; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; phosphodiesterase 8B; recurrent miscarriage; single nucleotide polymorphism; thyroid; guidelines; hypothyroidism; pregnancy; survey; thyroid testing; screening; iodine; iodine deficiency; median urinary iodine concentration; Obstetrik och gynekologi; Obstetrics and Gynaecology;

    Abstract : Hypothyroidism is a common endocrine disorder affecting women of reproductive age. On a global level, iodine deficiency is still the most common cause of hypothyroidism. Also genetic variations, in particular SNP rs4704397 in the PDE8B gene, are responsible for a significant proportion of TSH variations. READ MORE

  5. 5. Conformationally Constrained Nucleosides : Design, Synthesis, and Biochemical Evaluation of Modified Antisense Oligonucleotides

    Author : Oommen P. Varghese; Lars Engman; Jacek Stawinski; Uppsala universitet; []

    Abstract : This thesis is concerned with synthesis, structure and biochemical analysis of chemically modified oligonucleotides with potential therapeutic applications. The three types of chemical modifications described here are: (a) A North-East locked 1',2'-azetidine nucleoside (b) A North locked 2',4'-cyanomethylene bridged nucleoside and (c) A 2',4'-aza-ENA-T nucleoside.
