  1. 1. Benefits of Non-Linear Mixed Effect Modeling and Optimal Design : Pre-Clinical and Clinical Study Applications

    Author : Charles Ernest II; Andrew Hooker; Mats Karlsson; Bart Ploeger; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmacometrics; optimal design; nonlinear mixed effects models; population models; Pharmaceutical Science; Farmaceutisk vetenskap;

    Abstract : Despite the growing promise of pharmaceutical research, inferior experimentation or interpretation of data can inhibit breakthrough molecules from finding their way out of research institutions and reaching patients. This thesis provides evidence that better characterization of pre-clinical and clinical data can be accomplished using non-linear mixed effect modeling (NLMEM) and more effective experiments can be conducted using optimal design (OD). Abstract : Lung diseases continue to present a large burden to public health, especially in industrialized countries. For abetter understanding of the underlying patho-mechanisms in lung related diseases as well as for testing theefficacy of novel therapies, preclinical studies in animal models are indispensable. 3. 3. Change and variability in cognitive performance in old age. Effects of retest, terminal decline, and pre-clinical dementia

    Author : Valgeir Thorvaldsson; Göteborgs universitet; []
    Keywords : SAMHÄLLSVETENSKAP; SOCIAL SCIENCES; Aging; cognition; longitudinal; retest effects; terminal decline; pre-clinical;

    Abstract : The focus of the presented studies was to provide population estimates of change and variability in cognitive performance in old age. Data was drawn from the Gerontological and Geriatric Population Studies in Gothenburg, Sweden (H70). In study I retest effects were evaluated on level of performance in five cognitive abilities. 4. 4. Enhancing the therapeutic effect of biological drugs with protein engineering : Focusing on pre-clinical Alzheimer’s disease therapy

    Author : Fadi Rofo; Greta Hultqvist; Stina Syvänen; Sara Mangsbo; Joy Zuchero; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer’s disease; amyloid-beta; blood-brain barrier; transferrin receptor; protein-drugs; multivalent-bispecific antibodies;

    Abstract : Aggregation of the amyloid-β peptide (Aβ) is one of the main pathological hallmarks in Alzheimer’s disease (AD). The soluble Aβ aggregates (oligomers and protofibrils) have shown to be the most harmful species. Hence, targeting these aggregates can be of therapeutic potential. READ MORE
