  1. 1. Protein Folding, Binding and Evolution : PDZ domains and paralemmins as model systems

    Author : Greta Hultqvist; Per Jemth; Ruth Nussinov; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Protein folding; evolution; binding; allostery; Paralemmin; PDZ domain;

    Abstract : Proteins present at the synapse need to be multitasking in order to perform all vital functions in this limited space. In this thesis I have analyzed the function and evolution of such proteins, focusing on the PDZ domain and the paralemmin family. The PDZ domains bind to a wide variety of interaction partners. 2. 2. The Interaction Between the Chaperonin GroEL and Human Carbonic Anhydrase II : Protein Folding and Stability Studies

    Author : Malin Persson; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; carbonic anhydrase; protein folding; protein stability; chaperone; GroEL; folding mechanism; site-directed spinlabeling; electron paramagnetic resonance EPR ;

    Abstract : The presence of GroEL/ES during the refolding of human carbonic anhydrase II was found to increase the yield of active enzyme from approx. 70 to approx. 100%. This chaperone action on the enzyme could be obtained by adding GroEL alone, and the time course in that case was only moderately slower than the spontaneous process. READ MORE

  3. 3. Structure determination and thermodynamic stabilization of an engineered protein-protein complex

    Author : Elisabet Wahlberg; Per-Åke Nygren; Bengt-Harald Jonsson; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; affibody; protein structure; coupled folding; NMR spectroscopy; protein stability; protein-protein interactions; binding thermodynamics; isothermal titration calorimetry; protein engineering; Structural biochemistry; Strukturbiokemi;

    Abstract : The interaction between two 6 kDa proteins has been investigated. The studied complex of micromolar affinity (Kd) consists of the Z domain derived from staphylococcal protein A and the related protein ZSPA-1, belonging to a group of binding proteins denoted affibody molecules generated via combinatorial engineering of the Z domain. 4. 4. Studies on spinach plastocyanin and mutants : Expression in Escherichia coli, folding and function

    Author : Mikael Ejdebäck; Örjan Hansson; Miguel A. De la Rosa; Göteborgs Universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; plastocyanin; electron transfer; copper; protein-protein interactions; protein expression; folding; site-directed mutagenesis; NMR; Naturvetenskap; Natural sciences;

    Abstract : Photosynthesis is a process in which photons from sunlight excite chlorophylls in the thylakoid membranes of plants, algae and cyanobacteria. The photo-oxidised reaction centre chlorophyll P700 is re-reduced by an electron transferred from the soluble, blue copper protein plastocyanin. READ MORE

  5. 5. Post-synaptic Density Disc Large Zo-1 (PDZ) Domains : From Folding and Binding to Drug Targeting

    Author : Celestine Chi; Per Jemth; Maria Selmer; Sarah Linse; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; protein-protein interaction; protein folding; and drug design; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : Understanding how proteins fold and bind is interesting since these processes are central to most biological activity. Protein folding and protein-protein interaction are by themselves very complex but using a good and robust system to study them could ease some of the hurdles. READ MORE
