  1. 1. Studies of Eosinophil Cationic Protein (ECP) in vivo and in vitro : Impact of Genetic and Posttranslational Modifications

    Author : Jenny Eriksson; Per Venge; Agneta Trulson; Ester Boix; Uppsala universitet; []
    Keywords : Biomedicine; Eosinophil granulocyte; granular protein; ECP; gene polymorphism; cytotoxicity; SELDI-TOF MS; glycosylation; molecular and functional heterogeneity; fibrosis; Biomedicin;

    Abstract : Eosinophil granulocytes are tissue dwelling leukocytes that are implicated in host defence, particularly against helminthic parasites; they also participate in most inflammatory disorders. Although eosinophils have important roles in host defence mechanisms, their actions can also be harmful to the host as in the allergic inflammation where lung epithelium is destructed due to the release of toxic granule proteins.     Abstract : UDP-sugar metabolizing pyrophosphorylases provide the primary mechanism for de novo synthesis of UDP-sugars, which can then be used for myriads of glycosyltranferase reactions, producing cell wall carbohydrates, sucrose, glycoproteins and glycolipids, as well as many other glycosylated compounds. The pyrophosphorylases can be divided into three families: UDP-Glc pyrophosphorylase (UGPase), UDP-sugar pyrophosphorylase (USPase) and UDP-N-acety lglucosamine pyrophosphorylase (UAGPase), which can be discriminated both by differences in accepted substrate range and amino acid sequences.     Keywords : NATURVETENSKAP; NATURAL SCIENCES; Glycoconjugates; Carbohydrates; Galactose; Glucose; Solid-phase synthesis; SPOS; Glycosylation; Gel-phase 19F NMR spectroscopy; Fluorinated linker; Carbohydrate array; Microtiter plates; Carbohydrate-protein interactions; carbamate linker; Fsec; Protecting group; Fluorinated protecting group; Multivalent glycoconjugates; Adenovirus; Ad37; Epidemic keratoconjunctivitis; EKC; Sialic acid; Adenovirus inhibitor; Trivalent; Tetravalent; X-ray crystal structure; Bioorganic chemistry; Bioorganisk kemi; organisk kemi; Organic Chemistry;

    Abstract : To study the functions of glycoconjugates in biological systems reliable and efficient protocols for glycoconjugate synthesis are needed. To reach this goal we have developed methods for solid-phase synthesis of glycoconjugates that can be monitored with gel-phase 19F spectroscopy using fluorinated linkers, building blocks, and protecting groups. 4. 4. Genetic and serologic characterization of a Swedish human hantavirus isolate

    Author : Marie Lindkvist; Göran Bucht; Clas Alhm; Per Juto; Rainer Ulrich; Umeå universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; hantavirus; puumalavirus; diagnostics; HFRS; nucleocapsid protein; B-cell epitopes; epitope-mapping; protein interactions; glycosylation; antibody response; cross-reactivity; Virology; Virologi;

    Abstract : Hantaviruses are found practically all over the world and cause hemorrhagic fevers in man. Each year about 150,000 people are hospitalized in these zoonotic infections which can be of two types: hemorrhagic fever with renal syndrome (HFRS) or hantavirus cardiopulmonary syndrome (HCPS), depending on the infecting virus. Abstract : Binder-based assays are employed throughout the life sciences. Powerful signal amplification techniques have enabled detection of very rare molecule species diluted in simple buffers. Unspecific binding of primary binders leads to increased background in more complex samples. READ MORE
