  1. 11. Engineering membrane proteins for production and topology

    Author : Stephen Toddo; Daniel Daley; Roslyn Bill; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Membrane protein; Over-expression; Protein production; Codon optimization; Escherichia coli; AraH; NarK; mRNA secondary structure; coding sequence; ribosome binding site; RBS; topology; split-GFP; Biochemistry; biokemi;

    Abstract : The genomes of diverse organisms are predicted to contain 20 – 30% membrane protein encoding genes and more than half of all therapeutics target membrane proteins. However, only 2% of crystal structures deposited in the protein data bank represent integral membrane proteins. READ MORE

  2. 12. Metabolic Engineering of Recombinant Protein Production by Saccharomyces cerevisiae

    Author : Zihe Liu; Chalmers tekniska högskola; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; recombinant protein production; genome sequencing; DNA microarray; insulin precursor; growth rate; UV mutation; yeast; anaerobic electron acceptor; secretory pathway; unfolded protein response; metabolic engineering; systems biology; α-amylase;

    Abstract : The yeast Saccharomyces cerevisiae is a widely used cell factory for the production of fuels, chemicals, and it also provides a platform for the production of many heterologous proteins of medical or industrial interest. In this thesis, random and rational approaches, such as vector design, host engineering, fermentation analysis, UV Mutation, coupled with high-throughput systems biology techniques (including whole genomic sequencing, microarray analysis and flux analysis) and integrated analysis (Reporter feature technique), were employed to engineer cellular properties more effectively and purposefully to construct cell factories for protein production.     Keywords : NATURVETENSKAP; NATURAL SCIENCES; Escherichia coli; BL21 DE3 ; protein production; membrane proteins; secretory proteins; genome evolution; Biochemistry; biokemi;

    Abstract : The aim of my Ph.D. studies was to improve production yields of membrane- and secretory proteins in the widely used E. coli protein production strain BL21(DE3). READ MORE

  4. 14. Recombinant amelogenin - Strategies for purification and protein processing

    Author : Johan Svensson Bonde; Tillämpad biokemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Recombinant amelogenin; protein purification; fusion protein; purification tag; enamel; hydroxyapatite; matrix protein; dental regeneration; wound healing; bone formation;

    Abstract : Amelogenin is an extracellular matrix protein involved in formation of enamel during early tooth development. By interacting with the forming enamel crystallites amelogenin affects their growth, so that highly elongated hydroxyapatite crystals are formed. READ MORE

  5. 15. Structural Studies on PP2A and Methods in Protein Production

    Author : Auður Magnúsdóttir; Pär Nordlund; Herman Van Tilbeurgh; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; PP2A; PTPA; B56γ; Protein Production; Structural biology; Strukturbiologi; biokemi; Biochemistry;

    Abstract : PP2A is a major phosphatase in the cell that participates in multiple cell signaling pathways. It is a heterotrimer of a core dimer and variable regulatory subunits. Details of its structure, function and regulation are slowly emerging. Here, the structure of two regulators of PP2A are de-scribed; PTPA and B56γ. READ MORE
