  1. 21. Metabolic Engineering of Recombinant Protein Production by Saccharomyces cerevisiae

    Author : Zihe Liu; Chalmers tekniska högskola; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; recombinant protein production; genome sequencing; DNA microarray; insulin precursor; growth rate; UV mutation; yeast; anaerobic electron acceptor; secretory pathway; unfolded protein response; metabolic engineering; systems biology; α-amylase;

    Abstract : The yeast Saccharomyces cerevisiae is a widely used cell factory for the production of fuels, chemicals, and it also provides a platform for the production of many heterologous proteins of medical or industrial interest. In this thesis, random and rational approaches, such as vector design, host engineering, fermentation analysis, UV Mutation, coupled with high-throughput systems biology techniques (including whole genomic sequencing, microarray analysis and flux analysis) and integrated analysis (Reporter feature technique), were employed to engineer cellular properties more effectively and purposefully to construct cell factories for protein production. 2. 22. Exploring the interactome of EF-hand proteins

    Author : Mikael Bauer; Biofysikalisk kemi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; EF-hand; calcium; STIM; stromal interaction molecule; secretagogin; Calmodulin; calbindin D28k; protein-protein interactions; zinc; protein array;

    Abstract : EF-hand proteins are central in many cellular processes being the main group of Ca2+-sensing and buffering proteins in the cell. In the work presented in this thesis we study ligand interactions of three EF-hand proteins; calmodulin, calbindin D28k and secretagogin. READ MORE

  3. 23. Multiple functions of YopN in the Yersinia pseudotuberculosis type III secretion system : from regulation to in vivo infection

    Author : Sarp Bamyaci; Åke Forsberg; Edouard Galyov; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Type III secretion system; Yersinia; YopN; virulence; phagocytosis; mouse infection; secretion kinetics;

    Abstract : The type 3 secretion systems (T3SSs) are virulence mechanisms used by various Gram-negative bacteria to overcome the host immunity. They are often target-cell contact induced and activated. Activation results in targeting of virulence effector substrates into host cells. READ MORE

  4. 24. Regulation of glucagon secretion from pancreatic alpha cells

    Author : Yang De Marinis; diabetes och endokrinologi Genomik; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Epac2; glucagon secretion; pancreatic alpha cells; K- ATP channel; N-type Ca2 channel; L-type Ca2 channel; GLP-1; cAMP; PKA; Adrenaline; PKC;

    Abstract : Glucagon secreted by pancreatic α-cells plays an important role in the regulation of blood glucose. In this thesis, different techniques such as electrophysiology, immunohistochemistry and hormone secretion assay were combined to explore the mechanisms by which glucagon secretion is regulated. 5. 25. Membrane rafts and its GPI anchored complement inhibitors regulate insulin secretion

    Author : Vini Nagaraj; Diabetes - öpatofysiologi; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Membrane rafts; basal insulin secretion; regulated insulin secretion; CD59; CD55; SNARE proteins; insulin; pancreatic beta cells.;

    Abstract : Type 2 diabetic patients usually exhibit an abnormal lipid profile. One such lipid that is constantly elevated in T2D is cholesterol. At the cellular level, cholesterol aids in tight packing of sphingolipids in certain regions of the plasma membrane and these specialized regions are termed ‘membrane rafts’, which act as signalling hubs.
