  1. 21. Rate and Accuracy of Bacterial Protein Synthesis with Natural and Unnatural Amino Acids

    Author : Ka-Weng Ieong; Måns Ehrenberg; Simpson Joseph; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Ribosome; protein synthesis; translation; efficiency-accuracy trade-off; kinetics; unnatural amino acids; Molecular Biology; Molekylärbiologi;

    Abstract : This thesis addresses different questions regarding the rate, efficiency, and accuracy of peptide bond formation with natural as well as unnatural amino acids: Which step is rate-limiting during peptide bond formation? How does the accuracy vary with different transfer RNAs (tRNAs) and codons and how is it relevant to the living cells? Does proofreading selection of codon reading occur in a single- or multi-step manner as theoretically suggested? How does the E. coli translation system discriminate unnatural amino acids? Based on that, how to improve the incorporation efficiencies of unnatural amino acids?Based on the study on pH dependence of peptide bond formation, we show that the rate of the chemistry of peptidyl transfer to aminoacyl-tRNA (AA-tRNA) Gly-tRNAGly or Pro-tRNAPro limits the rate of peptide bond formation at physiological pH 7. 2. 22. Insights Into Protein Synthesis - Structural Aspects of Elongation Factor G and Fusidic Acid Inhibition

    Author : Sebastian Hansson; Biokemi och Strukturbiologi; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Molekylär biofysik; Structure chemistry; Strukturkemi; Molecular biophysics; Biologi; Biology; Biomedicinska vetenskaper; Biomedical sciences; conformational change; fusidic acid; protein synthesis; elongation factor G; crystal structures;

    Abstract : Elongation factor G (EF-G) catalyses the translocation step in protein synthesis on the ribosome. During this event the antibiotic Fusidic acid (FA) binds to and irreversably inhibits the release of the EF-G:GDP complex from the ribosome. READ MORE

  3. 23. Design, Synthesis and Characterization of Small Molecule Inhibitors and Small Molecule : Peptide Conjugates as Protein Actors

    Author : Jonas Nilsson; Lars Baltzer; Ingemar Kvarnström; Thomas Olsson; Linköpings universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Synthetic receptor; Peptide inhibitor conjugate; Peptide protein surface interactions; General Lead Discovery Library; Combinatorial Chemistry; Solid Phase Chemistry; Targeted Library; Thrombin inhibitor; Fluorescence; Surface Plasmon Resonance; Organic chemistry; Organisk kemi;

    Abstract : This thesis describes different aspects of protein interactions. Initially the function of peptides and their conjugates with small molecule inhibitors on the surface of Human Carbonic Anhydrase isoenzyme II (HCAII) is evaluated. READ MORE

  4. 24. Organization of mitochondrial gene expression in yeast : Specific features of organellar protein synthesis

    Author : Kirsten Kehrein; Martin Ott; Thomas A Fox; Stockholms universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Mitochondria; translation; bc1 complex; ribosome; gene expression; Biochemistry; biokemi;

    Abstract : Mitochondria contain their own genetic system, encoding key subunits of the oxidative phosphorylation system. These subunits are expressed by an organelle-specific gene expression machinery. READ MORE

  5. 25. Synthesis of Small Molecules Targeting ADP-Ribosyltransferases and Total Synthesis of Resveratrol Based Natural Products

    Author : Anders Lindgren; Mikael Elofsson; Nicholas Westwood; Umeå universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; organic synthesis; quinazolinone; ARTD; PARP; total synthesis; polyphenols; bensofuranes; organisk kemi; Organic Chemistry;

    Abstract : Diphtheria Toxin-like ADP-RibosyltransferasesThe Human ADP-ribosyl transferases (ARTDs) are a group of poorly studied enzymes which are believed to be involved in e.g. DNA repair, protein degradation, transcription regulation and cell death. Medicinal chemistry programmes aimed at developing selective inhibitors of these ARTDs were initiated. READ MORE
