21. Targeting HIV-1 entry and reverse transcription by vaccination
Abstract : Human immunodeficiency virus type I (HIV-1) is a complex retrovirus, which uses the CD4 receptor and chemokine receptors to infect its target cells. The chemokine receptor CCR5 is essential for primary HIV-1 infection. The hallmark of retroviruses is the enzyme reverse transcriptase (RT), which transcribes the virus genome from RNA to DNA. 22. Transcriptional and post-transcriptional regulation of telomerase reverse transcriptase (hTERT) expression : the role of histone modification and alternative splicing
Abstract : Telomerase, an RNA-dependent DNA polymerase responsible for telomeric DNA elongation, is silent in most normal differentiated human cells due to the tight repression of its catalytic unit, telomerase reverse transcriptase (hTERT), whereas constitutive hTERT expression and telomerase activation are prerequisites for cellular immortalization/malignant transformation. As reconfiguration of chromatin acts like a master on/off switch, determining whether particular genes are active or inactive, the project was specifically designed to first elucidate molecular strategies that integrate diverse signalling pathways at the hTERT chromatin and consequently result in telomerase activation or repression in normal and malignant human cells. 23. Biosensor Studies of Ligand Interactions with Structurally Flexible Enzymes : Applications for Antiviral Drug Development
Abstract : The use of a surface plasmon biosensor fills a missing link in kinetic studies of enzymes, since it measures directly the interaction between biomolecules and allows determination of parameters that are determined only indirectly in activity assays. The present thesis deals with kinetic and dynamic aspects of ligand binding to two viral enzymes: the human cytomegalovirus (HCMV) protease and the human immunodeficiency virus type 1 reverse transcriptase (HIV-1 RT). 24. Gene expression profiling with real-time PCR
Abstract : The sequence of the entire human genome is available today, and this information can be used to understand, characterize, and treat complex diseases such as cancer. Many cellular functions related to survival, growth and differentiation are reflected in the gene expression pattern, and the ability to quantify transcription levels of specific genes is, and always has been, central to research on gene function. 25. Computational Modelling of Ligand Complexes with G-Protein Coupled Receptors, Ion Channels and Enzymes
Abstract : Accurate predictions of binding free energies from computer simulations are an invaluable resource for understanding biochemical processes and drug action. The primary aim of the work described in the thesis was to predict and understand ligand binding to several proteins of major pharmaceutical importance using computational methods. READ MORE
