1. RNA in model lipid membranes - interactions in bulk and at surfaces
Abstract : The work presented in the thesis “RNA in model lipid membranes- interactions in bulk and at surfaces” aims to characterize the interaction between RNA and model membranes with different properties as well as to determine if ssRNA associate with model membranes in different ways. Several techniques have been used to characterize these systems, including quartz crystal microbalance with dissipation monitoring (QCM-D), Langmuir film balance in combination with neutron reflectometry and fluorescence microscopy as well as differential scanning calorimetry (DSC) and isothermal titration calorimetry (ITC). 3. Some Aspects of Physicochemical Properties of DNA and RNA
Abstract : This thesis is based on nine research publications (I – IX) on structure and reactivity of RNA vis-à-vis DNA. The DNA and RNA are made of flexible pentose sugar units, polyelectrolytic phosphodiester backbone, and heterocyclic nucleobases. 4. Studies on Nucleic Acids – Structure and Dynamics
Abstract : This thesis is based on six papers, Papers I-VI, focusing on the interplay between the stabilizing elements of nucleic acids self-assembly; hydrogen bonding, stacking and solvent effects. In Paper I we investigate how the substitution of the O4' for CH2 in the sugar moiety of adenosine (2'-deoxyaristeromycin) at the A6 position of the Dickerson-Drew dodecamer makes the two modified bases exist in a dynamic equilibrium between Hoogsteen and Watson-Crick base pairing in the NMR time scale.
