  1. 1. Studies on sterol 27-hydroxylase (CYP27A1)

    Author : Sara von Bahr; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Human sterol 27-hydroxylase (CYP27A1) has been studied in the present thesis with focus on substrate specificity and possible role of the enzyme for prevention of accumulation of cholesterol and cholestanol in tissues. Attempts have been made to evaluate a metabolic pathway to bile acids in human primary hepatocytes that starts with a 27-hydroxylation of cholesterol catalyzed by CYP27A1. READ MORE

  2. 2. Studies on human sterol 27-hydroxylase with emphasis on its mechanism of regulation and metabolic consequences of a deficient enzyme

    Author : Magnus Hansson; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Sterol 27-hydroxylase (CYP27A1) is a mitochondrial cytochrome P-450 enzyme present in most animal cells. In hepatocytes, CYP27A1 has an important role in connection with biosynthesis of bile acids from cholesterol. READ MORE

  3. 3. Cell death in atherosclerosis

    Author : Milita Crisby; Karolinska Institutet; Karolinska Institutet; []
    Keywords : atherosclerosis; carotid artery stenosis; ischemic stroke; apoptosis; necrosis; lipid peroxidation; plaque stability; pravastatin; inflammation; 27-[alpha]-hydroxylase;

    Abstract : Rupture of atherosclerotic lesions is recognized as the major cause of acute cerebrovascular and coronary syndromes. The characteristics of the typical vulnerable plaque include increased inflammatory cells at the shoulder region, a large lipid core, a thin fibrous cap with few collagen fibers and smooth muscle cells. 4. 4. Studies on a novel oxidative mechanism for elimination of extrahepatic cellular cholesterol

    Author : Amir H M Babiker; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : Recently, a new oxidative mechanism for elimination of cholesterol from extrahepatic cells by sterol 27-hydroxylase was described. This mechanism involves conversion of cholesterol into 27-hydroxycholesterol and cholestenoic acid. READ MORE

  5. 5. Use of primary human hepatocytes for the elucidation of bile acid synthesis

    Author : Ewa C S Ellis; Karolinska Institutet; Karolinska Institutet; []
    Keywords : ;

    Abstract : In the present investigation we cultured primary human hepatocytes on matrigel under serum-free conditions and studied bile acid formation. The following observations were made: · The major steroid products excreted from primary human hepatocytes into the culture medium were cholic acid, CA (70%) and chenodeoxycholic acid, CDCA (25%) conjugated with glycine or taurine.
