  1. 6. Exploring Selectivity and Hysteresis : Kinetic Studies on a Potato Epoxide Hydrolase

    Author : Diana Lindberg; Mikael Widersten; Gunnar Johansson; Richard N. Armstrong; Uppsala universitet; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; Epoxide hydrolase; regioselectivity; selectivity; hysteresis; conformational changes; cooperativity; deep eutectic solvents; methylstyrene oxide; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : The kinetic mechanism of an α/β hydrolase fold epoxide hydrolase from potato, StEH1, has been studied with the aims of explaining the underlying causes for enantio- and regioselectivity, both being important for product purity. Further effort has been laid upon understanding the causes of a hysteretic behavior discovered in the measurements leading to Paper I. READ MORE

  2. 7. Enzyme substrate solvent interactions : a case study on serine hydrolases

    Author : Linda Fransson; Karl Hult; Jürgen Pleiss; KTH; []
    Keywords : TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; lipase; esterase; specificity; Bioengineering; Bioteknik;

    Abstract : Reaction rates and selectivities were measured for transacylation of fatty acid esters in solvents catalysed by Candida antarctica lipase B and by cutinase from Humicola insolens. With these enzymes classical water-based enzymology can be expanded to many different solvents allowing large variations in interaction energies between the enzymes, the substrates and the surrounding. 3. 8. Lipase Specificity and Selectivity : Engineering, Kinetics and Applied Catalysis

    Author : Mohamad Takwa; Karl Hult; Bernhard Hauer; KTH; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Biokemi;

    Abstract : The specificity and selectivity of the enzyme Candida antarctica lipase B (CALB) were studiedfor several substrates and applications.With help of molecular modeling, the active site of CALB was redesigned for the ring openingpolymerization of D,D‐lactide. READ MORE

  4. 9. Enzyme selectivity as a tool in analytical chemistry

    Author : Anders Hamberg; Karl Hult; Harry Brumer; KTH; []
    Keywords : NATURVETENSKAP; NATURAL SCIENCES; substrate specificity; competing substrates; enantiomeric excess analysis; library screening; ladder sequencing; carboxypeptidase Y; 2-pyridylmethylamine; MALDI; Candia antarctica lipase B; pig liver esterase; horse liver alcohol dehydrogenase; acylated cyanohydrins; Biochemistry; Biokemi;

    Abstract : Enzymes are useful tools as specific analytical reagents. Two different analysis methods were developed for use in the separate fields of protein science and organic synthesis. Both methods rely on the substrate specificity of enzymes. Enzyme catalysis and substrate specificity is described and put in context with each of the two developed methods. READ MORE

  5. 10. Modulating Enzyme Functions by Semi-Rational Redesign and Chemical Modifications : A Study on Mu-class Glutathione Transferases

    Author : Malena A Norrgård; Bengt Mannervik; Birgitta Tomkinson; Timothy Mantle; Uppsala universitet; []
    Keywords : MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; protein redesign; semi-rational redesign; saturation mutagenesis; iterative saturation mutagenesis; chemical modification; Cys; Cys-X scanning; enzyme evolution; promiscuous; substrate selectivity; enantioselectivity; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Abstract : Today, enzymes are extensively used for many industrial applications, this includes bulk and fine-chemical synthesis, pharmaceuticals and consumer products. Though Nature has perfected enzymes for many millions of years, they seldom reach industrial performance targets. READ MORE
